摘要 目的探讨半乳糖凝集素9(Galectin-9)和转化生长因子β1(TGF-β1)在甲状腺乳头状癌(PTC)组织中的表达及其在PTC发生发展、浸润转移中的可能作用。方法选取信阳市中心医院2014年6月至2016年6月间甲状腺癌手术病例作为研究对象,PTC 108例和癌旁甲状腺组织35例,应用免疫组织化学方法检测其中Galectin-9和TGF-β1蛋白的表达情况,并分析二者与PTC患者临床病理特征的关系。结果 Galectin-9在PTC组织和癌旁组织中的阳性表达率分别为41.7%(45/108)、88.5%(31/35),其在PTC组织中的阳性表达率低于甲状腺癌旁组织(P<0.05);TGF-β1在PTC组织和癌旁组织中的阳性表达率分别为78.7%(85/108)、8%(3/35),其在PTC组织中的阳性表达率高于甲状腺癌旁组织,差异有统计学意义(P<0.05)。PTC组Galectin-9阳性表达率(41.7%)低于癌旁甲状腺组(88.5%),差异有统计学意义(P<0.05);PTC组TGF-β1阳性表达率(78.7%)高于癌旁甲状腺组(8%),差异有统计学意义(P<0.05)。Galectin-9在肿瘤>1 cm、伴有淋巴转移及临床分期Ⅲ+Ⅳ期的甲状腺癌组织中的表达率降低,TGF-β1在伴有淋巴转移及临床分期Ⅲ+Ⅳ期的甲状腺癌组织中的表达率升高,差异有统计学意义(均P<0.05)。结论在PTC组织中Galectin-9阳性表达低于癌旁组织,而TGF-β1阳性表达高于癌旁组织。Galectin-9的表达程度与肿瘤大小、淋巴转移、临床分期呈负相关,TGF-β1的表达程度与PTC的淋巴转移、临床分期呈正相关,提示二者可能参与PTC的恶性增殖和侵袭转移。 Objective To investigate the expression of Galectin-9 and transforming growth factor-β1(TGF-β1)in PTC and its possible role in the occurrence, development and invasion of PTC.Methods The cases of thyroid cancer admitted to Xinyang Central Hospital from June 2014 to June 2016 were selected as the research objects. A total of 108 cases of PTC and 35 cases of paracancerous normal thyroid tissue were selected, and the expression of galectin-9 and TGF-β1 proteins were detected by immunohistochemistry, the correlation between the two and the clinical pathological features of PTC patients were analyzed.Results The positive rate of Galectin-9 in PTC tissues and adjacent normal tissues were 41.7%(45/108) and 88.5%(31/35) respectively. The positive rate of Galectin-9 in PTC tissues was lower than that in adjacent thyroid carcinoma tissues(P<0.05). The positive expression rate of TGF-β1 in PTC tissues and adjacent normal tissues were 78.7%(85/108) and 8%(3/35) respectively. The positive expression rate of TGF-β1 in PTC tissues was higher than that in adjacent thyroid carcinoma tissues. The difference was statistically significant(P<0.05). The expression rate of Galectin-9 in thyroid cancer tissues with tumor >1 cm, lymph node metastasis and clinical stage Ⅲ+Ⅳ stage was significantly reduced. The expression rate of TGF-β1 increased significantly in thyroid cancer tissues with lymph node metastasis and clinical stage Ⅲ+Ⅳ stage. The differences were statistically significant(all P<0.05).Conclusion The expression of Galectin-9 in PTC tissues is significantly lower than that in adjacent normal tissues, while the expression of TGF-β1 is significantly higher than that in adjacent normal tissues. The expression level of Galectin-9 is negatively correlated with tumor size, lymph node metastasis and clinical stage. The expression level of TGF-β1 is positively correlated with lymph node metastasis and clinical stage of PTC. These results suggest that the two may participate in the malignant proliferation and invasion and metastasis of PTC.
