摘要 目的观察SOX12、CDK4在肝癌细胞(HepG2)与正常肝细胞(HB611)中的表达情况,分析SOX12与肝细胞癌的关系。方法采用逆转录-聚合酶链反应(RT-PCR)法检测HepG2、HB611细胞中SOX12 mRNA、CDK4 mRNA的表达水平,采用Western blot法检测HepG2、HB611细胞中SOX12蛋白、CDK4蛋白的表达水平,采用基因抑制剂shRNA抑制两种细胞中的SOX12基因,采用RT-PCR法检测两种细胞中SOX12 mRNA与CDK4 mRNA的表达情况,分析SOX12与肝细胞癌的关系。结果 SOX12 mRNA在HepG2细胞中表达水平高于HB611细胞,CDK4 mRNA在HepG2细胞中表达水平低于HB611细胞,差异有统计学意义(均P<0.05)。两种细胞中SOX12蛋白表达水平比较,差异无统计学意义(P>0.05),CDK4蛋白在HB611细胞中的表达水平高于HepG2细胞,差异有统计学意义(P<0.05)。抑制SOX12基因后,两种细胞SOX12 mRNA的表达水平较抑制前降低,CDK4 mRNA表达水平也较抑制前降低,差异有统计学意义(均P<0.05)。结论 SOX12与CDK4在HepG2、HB611细胞中均有表达,在两种细胞中SOX12 mRNA、CDK4 mRNA的表达水平存在差异,SOX12蛋白表达水平相近,CDK4蛋白表达水平存在差异。SOX12可能通过影响CDK4的表达参与肝细胞癌的发展进程,可能成为治疗与预测肝细胞癌预后新的标志物。 Objective To observe the expression of SOX12 and CDK4 in hepatocellular carcinoma cells(HepG2) and normal hepatocytes(HB611). The relationship between SOX12 and hepatocellular carcinoma was analyzed.Methods The expression levels of SOX12 mRNA and CDK4 mRNA in HepG2 and HB611 cells were detected by reverse transcription-polymerase chain reaction(RT-PCR). Western blot was used to detect the expression levels of SOX12 protein and CDK4 protein in HepG2 and HB611 cells. Gene inhibitor shRNA was used to inhibit SOX12 gene in the two kinds of cells, and the expression levels of SOX12 mRNA and CDK4 mRNA in the two kinds of cells were detected by RT-PCR, and the relationship between SOX12 and hepatocellular carcinoma was analyzed.Results The expression level of SOX12 mRNA in HepG2 cells was higher than that in HB611 cells, and the expression level of CDK4 mRNA in HepG2 cells was higher than that in HB611 cells, and the differences were statistically significant(both P<0.05). The expression levels of SOX12 protein in two kinds of cells were compared, and the difference was not statistically significant(P>0.05). The expression level of CDK4 protein in HB611 cells was higher than that in HepG2 cells, and the difference was statistically significant(P<0.05). After inhibiting SOX12 gene, the expression levels of SOX12 mRNA in the two cells were lower than those before inhibiting, and the expression levels of CDK4 mRNA were also lower than those before inhibiting, and the differences were statistically significant(all P<0.05).Conclusion SOX12 and CDK4 were both expressed in HepG2 and HB611 cells. The expression levels of SOX12 mRNA and CDK4 mRNA in the two cells were different, and the expression levels of SOX12 protein were similar, and the expression levels of CDK4 protein were different. SOX12 may participate in the development of hepatocellular carcinoma by influencing the expression of CDK4, which may become a new marker for treatment and prediction of prognosis of hepatocellular carcinoma.
