摘要
目的探讨拳参中总黄酮粗品及精品的体外抑菌作用。方法采用乙醇回流提取拳参总黄酮,HPD-400型大孔吸附树脂进行纯化,通过Kirby-Baue纸片扩散法研究总黄酮粗品及精品对金黄色葡萄球菌、大肠杆菌的抑制效果,并采用二倍稀释法测定了最低抑菌浓度(MIC)。结果拳参总黄酮粗品对金黄色葡萄球菌、大肠杆菌的最低抑菌浓度分别为0.2815mg/ml、0.2815mg/ml。拳参总黄酮精品对金黄色葡萄球菌、大肠杆菌的最低抑菌浓度分别为0.08257mg/ml、0.08257mg/ml。结论拳参总黄酮粗品和精品对金黄色葡萄球菌、大肠杆菌均有一定的抑制作用,通过对比发现,总黄酮精品对2种菌抑制作用强于总黄酮粗品,也说明拳参中黄酮类成分发挥了抑制作用,但具体是哪种黄酮类成分还有待进一步研究。
Objective: To study the in vitro antibacterial activity of the crude and fine extract of total flavonoids from Polygonum bistorta L. METHODS Ethanol extraction and HPD-400 macroporous resins for purification were selected. The content of total flavonoids in Polygonum bistorta L.were determined by UV spectrophotometry with rutin as standard control. The antimicrobial activity of total flavonoids in Polygonum bistorta L. to Eseheriehia coli and Staphylococcus aureus was investigated through paper diffusion method and double dilution method.RESULTS The minimum inhibitory concentrations the crude extract of total flavonoids were showed as follows: 0.2815mg/ml to Staphylococcus aureus and 0.2815 mg/ml to Eseheriehia coli. The minimum inhibitory concentrations the fine extract of total flavonoids were showed as follows: 0.08257mg/ml to Staphylococcus aureus and 0.08257 mg/ml to Eseheriehia coli. Conclusion The crude and fine extract of total flavonoids from Polygonum bistorta L. showed the strong antimicrobial effects for bacterium. The fine extract of total flavonoids has a certain effect on the two kinds of bacteria,but the main ingredient of total flavonoids need further study.
