摘要
目的 探讨C反应蛋白(hs-CRP)-757T>C基因变异与缺血性脑卒中发生的相关性,为揭示CRP在缺血性脑卒中发生过程中的作用提供理论依据。方法 从2016年3月至2017年5月,共有155例缺血性脑卒中患者纳入该研究,并选取同期体检健康者120例作为对照。常规抽取外周血3mL,提取DNA后采用聚合酶链式反应(PCR)法扩增后直接测序检测C反应蛋白-757T>C基因位点的多态性,比较两组CRP-757T>C基因型发生的频率、不同等位基因的分布,分析CRP-757T>C基因多态性与缺血性脑卒中的关系。结果 脑卒中组hs-CRP、TG、TC浓度显著高于对照组,差异有统计学意义(P<0.05)。Logistic回归分析发现,高血压、糖尿病、冠心病及hs-CRP均为缺血性脑卒中发生的独立因素,且CRP-757T>CCC纯合子或-757C等位基因频率(21.29%和36.45%)显著高于对照组(12.50%和27.92%),且与hs-CRP浓度显著相关(P<0.05)。结论 CRP-757T>C基因多态性可能会影响血清hs-CRP的水平,且与缺血性脑卒中有关。
Objective To explore the relationship between C reactive protein (CRP) gene polymorphisms -757 T>C and susceptibility to ischemic stroke,and provide theoretical basis for the effect of -757 T>C on the occurrence of ischemic stroke. Methods From March 2016 to May 2017,155 cases of ischemic stroke patients were selected as experimetal group,and 120 cases of healthy people were also recruited as control group.Peripheral blood of each participant was collected and genomic DNA was extracted,CRP -757T>C genotypes were detected.The relationship between CRP -757T>C genotypes and ischemic stroke were analysed Results Compared with control group,the levels of hs-CRP,TG,TC were higher ( P <0.05).The results of logistic regression analysis showed that hypertension,diabetes mellitus,coronary heart disease and hs-CRP were all the independent factor in the occurrence of ischemic stroke.The frequency of -757 T>C CC or C-allele are higher than the control group,which are correlated with the level of hs-CRP( P <0.05). Conclusion CRP -757 T>C genotypes might influence the level of hs-CRP,which might be related with ischemic stroke.
