摘要
目的:通过检测胃腺癌及癌旁组织中CD133、LGR5的表达情况,分析两者在胃腺癌中的表达及其与临床病理参数之间的关系,探讨两者在胃腺癌发生、发展过程中的意义。方法:采用MaxVision免疫组化染色方法观察120例胃腺癌及相应癌旁组织中CD133及LGR5的表达,分析它们与各临床病理参数之间的关系。结果:⑴CD133在胃腺癌及癌旁组织中的阳性率分别为36. 7%及5%,二者中CD133的表达差异有统计学意义(P <0. 05);对比胃腺癌中CD133表达在不同Lauren分级、脉管瘤栓、淋巴结转移及临床分期间的差异,P均<0. 05,差异有统计学意义。⑵LGR5在胃腺癌及癌旁组织中的阳性率分别为30. 8%及2. 5%,二者中LGR5的表达差异有统计学意义(P <0. 05);对比胃腺癌中LGR5表达在不同脉管瘤栓、淋巴结转移及临床分期间的差异,差异有统计学意义(P <0. 05)。⑶胃腺癌组织中CD133与LGR5的表达存在正相关(r=0. 278,P <0. 05)。结论:肿瘤干细胞标记物CD133及LGR5在胃腺癌的发生、侵袭、转移中发挥重要作用;CD133与LGR5之间存在正相关,提示两者在胃腺癌的发生及发展过程中可能存在协同作用。
Objective: To study the expression and clinical significance of CD133 and LGR5 in gastric adenocarcinoma and adjacent tissues, and to explore their roles in the occurrence and onset of gastric adenocarcinoma. Methods: The expressions of CD133 and LGR5 in 120 cases of gastric adenocarcinoma and corresponding adjacent tissues were detected by MaxVision immunohistochemical staining. The relationship between CD133 and LGR5 with every pathologic parameters of gastric adenocarcinoma was analyzed statistically. Results:(1)The positive rates of CD133 in gastric adenocarcinoma and adjacent tissues were 36.7% and 5%, and the difference was statistically significant ( P <0.05). The expression of CD133 protein was associated with Lauren classification, vascular tumor thrombus, lymph node metastasis and clinical stage in patients with gastric adenocarcinoma ( P <0.05).(2)The positive rates of LGR5 in gastric adenocarcinoma and adjacent tissues were 30.8% and 2.5%, and the difference was statistically significant ( P <0.05). The expression of LGR5 protein was associated with vascular tumor thrombus, lymph node metastasis and clinical stage in patients with gastric adenocarcinoma ( P <0.05).(3)There was a positive correlation between the expression of CD133 and LGR5 in gastric adenocarcinoma tissues ( r =0.278, P <0.05). Conclusions: Cancer stem cell markers CD133 and LGR5 play an important role in the occurrence, invasion and metastasis of gastric adenocarcinoma.There is a positive correlation between CD133 and LGR5, suggesting that there may be synergistic effects in the occurrence and development of gastric adenocarcinoma .
