托吡卡胺的毛细管电泳拆分

摘要 目的 利用毛细管电泳法拆分抗胆碱类手性药物托吡卡胺。方法 利用毛细管电泳法,筛选托吡卡胺溶液质量浓度、波长、缓冲液浓度等条件,添加磺酸化β-环糊精聚合物(S-β-CDP),拆分托吡卡胺。结果 托吡卡胺最佳拆分条件:托吡卡胺质量浓度5mg/L、波长254nm、进样时间10s、磷酸盐缓冲液浓度30mmol/L、磷酸盐缓冲液pH值4.5、S-β-CDP质量浓度30mg/L、S-β-CDP与磷酸盐缓冲液比例1∶1,加入S-β-CDP前分离度为7.9,加入S-β-CDP后分离度为8.139。结论 以S-β-CDP为手性添加剂,采用毛细管电泳法拆分手性药物托吡卡胺,可以实现良好拆分。 Objective To separate the anticholinergic drug tropicamide by capillary electrophoresis. Methods By capillary electrophoresis method,screening of tropicamide solution mass concentration,wavelength,concentration of phosphate buffer,etc.,and add sulfonated beta-cyclodextrin polymer(S-β-CDP)to split tropicamide. Results The best separation condition was:5 mg/L tropicamide,the wavelength of 254 nm,sample quantity for 10 s,concentration of phosphate buffer for 30 mmol/L,phosphate buffer solution pH value of 4.5,S-β-CDP concentration of 30 mg/L,S-β-CDP and phosphate buffer ratio of 1∶1.And tropicamide separation degree was 7.9 before the S-β-CDP,while tropicamide separation degree was 8.139 after the S-β-CDP. Conclusion In this study,S-β-CDP was used as a chiral additive,and by capillary electrophoresis,tropicamide could be well separated.
