促生长激素神经肽对坐骨神经损伤后血管内皮生长因子表达的影响

摘要 目的研究促生长激素神经肽(Gal)对大鼠坐骨神经损伤后血管内皮生长因子(VEGF)表达的影响。方法共选取36只健康雄性SD大鼠,将其随机分为正常组(A组)、对照组(B组)、促生长激素神经肽组(C组),每组大鼠12只。A组不做任何处理,B、C组造成大鼠坐骨神经SeddonⅡ度损伤模型,24h后进行实验,A组正常饮食、不进行任何治疗,B组加强营养、不进行任何治疗,C组进行注射Gal3μg/d治疗,每天1次,共4周。分别于治疗2周和4周后,采用免疫组化染色方法,检测大鼠血管内皮生长因子(VEGF)的表达。结果2周时,B、C组VEGF达到最高值,4周时开始下降,且B、C组两两比较,VEGF在2周和4周时均有增高。结论促生长激素神经肽可以使坐骨神经损伤后修复加快,同时坐骨神经损伤后的修复机制可能与血管内皮生长因子(VEGF)的表达有关。 Objective To Study the galanin on vascular injury endothelial growth factor(VEGF)expression effects after rat sciatic nerve.Methods A total of thirty-six healthy male SD rats,were randomly divided into normal group(A group),control group(B group),galanin group(C group),12 rats in each group.A group without any treatment,B and C group of rats were caused by sciatic nerve injury model of SeddonⅡ.Rats began experimenting after 24 hours,A group was fed with normal diet without any treatment,B group strengthened nutrition without any treatment,C group was injected with galanin 3μg/d.Therapy time was a time a day,for 4 weeks.After 2 weeks and 4 weeks,we used rat footprinting method,immunohistochemical method to understand the rat sciatic nerve function index(SFI)score,and detect rat vascular endothelial growth factor(VEGF)expression.Results At 2 weeks,VEGF of B,C group reached the highest value,and began to decline at 4 weeks.B,C group VEGF at 2 weeks and 4 weeks has increased respectively.Conclusion Galanin can accelerate repair of sciatic nerve injury,and repair mechanism after sciatic nerve injury may be related to vascular endothelial growth factor(VEGF)expression.
