抵当汤减钩吻毒性的实验研究

Experimental Study on Reducing the Detoxification Function of Didang Decoction to Gelsemium

目的观察抵当汤对钩吻的解毒作用。方法通过预实验确认钩吻致死量后,采用传统水煎煮和乙醇提取方式的抵当汤与钩吻,通过不同给药方式的毒性试验检测小鼠死亡情况;给药后小鼠眼球后静脉丛取血和肺组织取材,观察各组血常规、乙酰胆碱酯酶变化以及小鼠肺组织HE染色情况。结果急性毒性预实验结果显示:钩吻的Dm剂量为1.5g/kg。急性毒性实验结果显示:钩吻配伍抵当汤后,小鼠死亡率均明显下降,且死亡时间亦明显延长(P<0.05)。与空白组相比,钩吻组小鼠RBC、PLT、HGB、ACH与空白组比较呈显著性差异(P<0.05),抵当汤钩吻混合液组小鼠RBC、PLT、HGB、ACH与钩吻组比较呈显著性差异(P<0.05),而与空白组无差异。HE染色显示:钩吻抵当汤混合给药后小鼠肺组织细支气管、肺泡壁、肺间质中性粒细胞浸润伴充血减轻,肺泡壁毛细血管扩张不明显,肺泡腔渗出减少,肺组织局灶性实变消失。结论抵当汤能减缓钩吻毒性。

Objective: Observe the detoxification function of Didang Decoction to Gelsemium. Methods: Didang Decoction is made by adopting the traditional water-decocting method and ethanol-extracting method. Gelsemium is made by ethanol-extracting method. Mice were feed Didang Decoction and Gelsemium by different order. Draw blood from the venous plexus posterior to eyeballs of the subjects and material from lung tissue, observe the changes in blood routines of respective groups, acetylcholinesterase and HE stained preparation for the lung tissue of the mice. Results: The result of pre-experiment of acute toxicity shows the DM dose of Gelsemium is 1.5 g/kg;The experimental result of acute toxicity shows the compatibility of Gelsemium and Didang Decoction decreses the death rate of mice and the post-mortem interval has been conspicuously prolonged(P<0.05). Comparing with the control group, the RBC、PLT、HGB、ACH of the mice in the group taking in Gelsemium showed a significant difference(P<0.05). Comparing with the group taking in Gelsemium, the RBC, PLT, HGB, ACH of the mice in the group taking Didang Decoction and Gelsemium at the same time show a significant difference(P<0.05). There was show no difference with blank control group. HE Staining shows after taking in Didang Decoction and Gelsemium at the same time, the infiltration and hyperemia of lung tissue bronchiole, alveolar wall, lung interstitial neutrophil cell are alleviated, capillary of alveolar showed no conspicuous hemangiectasis, exudation from alveolar were reduced and local lesions of lung tissue disappear. Conclusion: Didang Decoction can detoxify Gelsemium toxicity.
