三七总皂苷对大鼠腹膜纤维化模型氧化应激水平的影响研究 被引量:2

摘要 目的通过建立腹膜纤维化大鼠模型,探讨三七总皂苷(PNS)对腹膜纤维化大鼠模型氧化应激的影响。方法20只SD健康大鼠随机分为空白组(5只)、模型组(8只)和试验组(7只)。空白组每日腹腔内注射生理盐水(100ml/kg),1次/d;模型组每日腹腔内注射4.25%葡萄糖腹透液(100ml/kg),同时隔日联合脂多糖1.2mg/kg腹腔注射(第1、3、5……天),1次/d;试验组每日腹腔内注射4.25%葡萄糖腹透液100ml/kg与PNS(35mg/kg)混合液,同时隔日联合脂多糖1.2mg/kg腹腔注射(第1、3、5……天)。透析28d后,取大鼠壁层腹膜做病理切片观察。检测并比较3组大鼠血清中晚期糖基化终末产物(AGEs)、还原型谷胱甘肽(GSH)、丙二醛、总超氧化物歧化酶(T-SOD)的含量。结果光镜下观察壁层腹膜病理切片,模型组大鼠腹膜组织明显增厚,并可见炎症细胞浸润、纤维细胞及血管增生明显增多,而试验组上述改变明显改善。试验组与空白组比较,AGEs、丙二醛明显增高,T-SOD明显减少,差异有统计学意义(P<0.05);模型组与空白组比较,AGES、丙二醛明显增加,T-SOD、GSH降低,差异均有统计学意义(均P<0.05);试验组与模型组比较,AGEs明显降低,差异有统计学意义(P<0.05)。结论隔日腹腔注射1.2mg/kg脂多糖,可成功建立腹膜纤维化大鼠模型;PNS可降低AGEs含量,改善腹膜功能。 Objective To explore the effect of Panax notoginseng saponins (PNS) on oxidative stress in peritoneal fibrosis rat model by establishing peritoneal fibrosis rat model.Methods Totally 20 healthy rats were randomly divided into control group( n =5),model group( n =8) and experimental group( n =7).The control group was given intraperitoneal injection of normal saline(100 ml/kg),once a day;the model group was given intraperitoneal injection of 4.25% dialysate(100 ml/kg,once a day) combined with lipopolysaccharide(LPS);while the experimental group was given intraperitoneal injection of 4.25% dialysate and PNS(35mg/kg),combined with LPS.After 28 days,the parietal peritoneu was to be done pathological observation.The advanced glycationend products(AGEs),glutathione(GSH),malondialdehyde(MDA) and superoxide dismutase(SOD) in serum of three groups were detected and compared.Results Observing under light microscope,rat peritoneum tissues in the model group were significantly thickened,inflammatory cell infiltration,fibrocyte and vascular hyperplasia were increased significantly.However,the above changes in the experimental group were improved obviously.Compared with the control group,there were significant increases in AGEs and MDA,and significant decreases in T-SOD in experimental groupwith statistically significant differences ( P <0.05 for each );compared with the control group,there were significant increases in AGEs and MDA,and significant decreases in T-SOD and GSH in model group with statistically significant differences ( P <0.05 for each);compared with the model group,there were significant decreases in AGEs in experimental group with statistically significant difference ( P <0.05).Conclusion Peritoneal fibrosis rat model can be successfully established by intraperitoneal injection of 1.2 mg/kg LPS once every other one day;PNS can make the level of AGEs reduced,the level of T-SOD,GHS raised and experimental fibrosis eased,thus peritoneal function were be improved.
