摘要 目的建立并评价抽动秽语综合征SD大鼠自身免疫动物模型的有效性。方法收集难治性抽动秽语综合征患者血清并检测血清中的抗核抗体浓度,将30只SD大鼠随机分为模型组、低剂量对照组和空白对照组,将抗核抗体含量高的血清、含量低的血清和PBS液分别通过微泵缓慢注射到3组大鼠的纹状体内,记录和观察大鼠的行为变化并进行分析。结果共收集了20名抽动秽语综合征患者的血清,血清抗核抗体平均浓度为(9.25±7.65)μg/ml,最高为24.47μg/ml,模型组从注射血清次日开始出现刻板症状,在第3~5天达到高峰,刻板行为评分高于低剂量对照组和空白对照组(P<0.01)。结论含高浓度抗核抗体的难治性抽动秽语综合征患者血清注入大鼠纹状体内可成功复制出抽动秽语综合征的特征性抽动症状,为抽动秽语综合征的临床以及基础研究提供了有效的工具。 Objective To establish and evaluate the effectiveness of the animal model of tourette syndrome(TS)in Sprague-Dawley rats. Methods Thirty Sprague-Dawley rats were randomly divided into model group, low dose control group and blank control group. Serum with high antinuclear antibody content, serum with low antinuclear antibody content and PBS solution were slowly injected into the striatum of two or three groups of rats by micropump, respectively. The behavior changes of rats were recorded, observed and analyzed. Results Serum samples from 20 tourette syndrome patients were collected. The concentration of serum ANA was(9.25±7.65)μg/ml and the highest was 24.47 μg/ml. The model group began to show stereotypic symptoms the next day after injection of serum and reached the peak on the 3rd to 5th day. The score of stereotypic behavior was higher than that of low dose control group and blank control group.(P<0.01). Conclusion The characteristic tic symptoms of tourette syndrome was successfully reproduced by injecting the serum of patients with refractory Tourette syndrome with high concentration of anti-nuclear antibody into the striatum of rats, which provided an effective tool for clinical and basic research of Tourette syndrome.
