淀粉样前体蛋白在结肠癌患者组织中的表达水平及其与预后的相关性研究

Expressions of amyloid precursor protein in colon cancer tissues and their correlations with prognosis

目的探讨淀粉样前体蛋白(amyloid precursor protein,APP)在结肠癌患者癌组织与癌旁组织中的表达差异,并初步验证该差异与患者预后之间的相关性。方法收集桂林市人民医院手术切除并由病理科组织学检查确诊的结肠癌及癌旁正常的组织各30例。通过实时荧光定量PCR实验和蛋白印迹实验分别在转录水平和翻译水平检测APP在癌组织与癌旁组织中的表达。同时结合TCGA数据库分析APP的表达与患者预后的相关性。结果结肠癌组织中APP的mRNA和蛋白表达水平显著高于癌旁组织(P<0.05)。同时Kaplan-Meier生存分析证实APP低表达的结肠癌患者60个月生存期显著高于APP高表达的患者(P<0.05)。结论APP在结肠癌组织中高表达,并与结肠癌患者的预后呈负相关,提示APP可能是结肠癌患者预后的重要参考指标。

Objective To explore the difference of expressions of amyloid precursor protein(APP)in cancer tissues and paracancerous tissues of colon cancer patients, and to verify the correlations between this difference and patient prognosis. Methods Thirty samples of colon cancer and normal tissues adjacent to the tumors were collected from the hospital for surgical resection and pathological section. The expressions of APP in cancer tissues and adjacent tissues were detected by qRT-PCR and Western blotting at the transcriptional and translational levels, respectively. At the same time, the TCGA database was used to analyze the correlations between APP expressions and patient prognosis. Results The mRNA and protein expression levels of APP in colon cancer tissues were significantly higher than those in normal tissues(P<0.05). At the same time, Kaplan-Meier analysis confirmed that the rate of 60 months survivalof patients with low expressions of APP was significantly higher than that of patients with high expressions of APP(P<0.05). Conclusion APP is highly expressed in colon cancer tissues and highly correlated with the prognosis of colon cancer patients. The above results suggest that APP may be an important index for the prognosis of colon cancer patients.
