摘要
目的建立实时荧光PCR检测鸭血中鸭、猪、牛等动物源性成分的方法。方法对重庆市火锅店销售的鸭血进行采样检测。提取鸭血样品DNA,进行实时荧光PCR检测,确定循环阈值,分析样品中鸭、猪、牛源性成分。结果DNA提取液OD260nm/OD280nm值为1.8~2.0,纯度较高;8个市售鸭血样品中2个样品只检出猪源性成分,1个样品同时检出鸭源性成分和猪源性成分,研究发现所采集的鸭血存在猪血掺假的可能。结论该方法快捷、可操作性强,适用于鸭血中动物源性成分的测定。
Objective To establish a method for the detection of animal-derived ingredients including duck, pig and cow in duck blood by real-time fluorescence PCR. Methods Samples of duck blood were taken from hot pot restaurants in Chongqing. DNA was extracted from duck blood curd samples and detected by real-time PCR. The cycle threshold was determined, and the duck, porcine and bovine-derived ingredients were analyzed. Results OD260 nm/OD280 nm of DNA extract was 1.8-2.0, and the purity was high. Porcine-derived ingredients were detected in 2 samples out of 8 duck blood curds, and both duck and porcine-derived ingredients were detected in 1 sample. It was found that the collected duck blood might be adulterated with pig blood. Conclusion This method is rapid and operable, which is suitable for the determination of animal-derived components in duck blood.
