摘要
目的 观察盐霉素(SAL)联合表没食子儿茶素没食子酸酯(EGCG)对人胃癌细胞株SGC-7901增殖、凋亡能力的影响,并探讨其机制。方法 取对数生长期SGC-7901细胞并分组,联合组分别加入5、10、20、40mg/L的EGCG及2μmol/L的SAL,SAL组加入1、2、4、8μmol/L的SAL,EGCG组加入5、10、20、40mg/L的EGCG,对照组无药物、只加培养液,比较各组药物作用24、48、72h后细胞增殖抑制率。取对数生长期SGC-7901细胞并分组,联合组加入2μmol/L的SAL和10mg/L的EGCG,SAL组加入2μmol/L的SAL,EGCG组加入10mg/L的EGCG,对照组无药物、仅加培养基,比较各组药物作用48h后荧光染色情况及bax、bcl-2mRNA表达情况。结果 SAL、EGCG单药均能抑制SGC-7901细胞增殖,联合用药效果更显著(P<0.05);对照组呈均匀绿色荧光,SAL组和EGCG组出现红色荧光,联合组红色荧光更明显;与对照组比较,联合组baxmRNA相对表达量降低,bcl-2mRNA相对表达量增加(P均<0.05)。结论 SAL、EGCG均能抑制SGC-7901细胞增殖,并诱导凋亡,两者联合应用效果更佳,机制可能与两者可降低bax表达及升高bcl-2表达有关。
Objective To investigate the effects of salinomycin (SAL) combined with epigallocatechin gallate (EGCG) on proliferation and apoptosis of human gastric cancer SGC-7901 cells and to explore its mechanism. Methods The SGC-7901 cells in the logarithmic phase were selected and divided into four groups;the cells in the combination group were added with 5, 10, 20, and 40 mg/L EGCG and 2 μmol/L SAL, the SAL group with 1, 2, 4, and 8 μmol/L SAL, the EGCG group with 5, 10, 20, and 40 mg/L EGCG, while the control group had no cells and was only added with the culture medium. The inhibitory rates of proliferation of SGC-7901 cells were compared at 24, 48 and 72 h after treatment. The SGC-7901 cells in the logarithmic phase were selected and divided into four groups;the cells in the combination group were added with 2 μmol/L SAL and 10 mg/L EGCG, the SAL group with 2 μmol/L SAL, the EGCG group with 10 mg/L EGCG, and the control group only with medium. The fluorescence staining and Bax, Bcl-2 mRNA expression of SGC-7901 cells in each group were detected at 48 h after treatment.Results SAL and EGCG alone could inhibit the proliferation of SGC-7901 cells, and the effect of combined use of SAL and EGCG was more significant ( P <0.05). The control group showed uniform green fluorescence, and red fluorescent apoptotic cells appeared in the two single-drug experimental groups, and the proportion of red fluorescent apoptotic cells was more obvious in the combined experiment. Compared with the control group, the relative expression of Bax mRNA decreased and the relative expression of Bcl-2 mRNA increased in the combination group ( P <0.05).Conclusions SAL and EGCG alone can inhibit the proliferation of SGC-7901 cells and induce apoptosis, and the effect of the combination of the two groups is better. The mechanism may be related to the decrease of Bax expression and the increase of Bcl-2 expression.
