摘要
目的探讨靶向γ-连接素(γ-catenin)基因的RNA干扰(RNAi)对人急性早幼粒细胞白血病细胞株HL-60细胞增殖的影响,为白血病的治疗寻找新的靶点。方法针对γ-catenin基因设计dsRNA,转染HL-60细胞,运用RT-PCR检测细胞γ-catenin mRNA表达,Westernblot检测γ-catenin蛋白表达,MTT法检测细胞增殖活性。结果经dsRNA转染后,γ-catenin基因表达及蛋白表达水平下降,HL-60细胞增殖受抑制。结论靶向γ-catenin基因的RNAi可以有效地降低HL-60细胞中γ-catenin基因表达,抑制HL-60细胞增殖,故γ-catenin基因可能作为潜在的白血病治疗靶点。
Objective To investigate the effect of RNA interference(RNAi) on proliferation in human acute promyelocytic leukemia HL-60 cell lines,and to find new targets for the treatment of leukemia. Methods In view of the γ-catenin gene design dsRNA,transfection HL-60 cells,using RT-PCR detection cells γ-catenin gene mRNA expression,Westernblot detect γ-catenin protein expression,determined by MTT method to detect cell proliferation activity. Results After dsRNA transfection,protein and gene expression levels of γ-catenin and the proliferation rate of HL-60 cells decreased. Conclusion RNAi targeted to the γ-catenin gene can effectively reduce the expression of γ-catenin gene in HL-60 cells,which can inhibit the proliferation of HL-60 cells,and γ-catenin gene can be used as a target for potential leukemia treatment.
