Research progress of single cell sequencing in the diagnosis and treatment of hematological diseases

摘要 目前广泛应用的二代测序(NGS)技术,是基于整个细胞群体的分析,不能观察细胞的异质性.单细胞测序(single cell sequencing,SCS)则是在单个细胞水平上,对基因组或转录组进行扩增并测序,以检测单核苷酸位点变异(SNV)、基因拷贝数变异(CNV)、单细胞基因组结构变异、基因表达水平、基因融合、单细胞转录组的选择性剪切、单细胞表观基因组的DNA甲基化状态等[1].SCS可实现在单细胞水平对疾病和生物学过程中的遗传学特征进行研究,包括早期胚胎发育、肿瘤发生和发展的分子学机制、肿瘤的异质性和进化过程、循环肿瘤细胞(CTC)和克隆演变等[2-4].
