摘要
目的:研究不同浓度肉苁蓉正丁醇萃取物对阿霉素造人正常胚肾上皮细胞株(HKC)细胞损伤的影响。方法:用不同浓度肉苁蓉的正丁醇萃取物,分别在不同时间作用于阿霉素(Dox)造HKC细胞体外损伤模型,并设空白对照和阴性对照,采用MTT比色法测定细胞存活率;采用流式细胞仪碘化丙啶(PI)单染法检测细胞周期分布。结果:阴性对照组与空白对照组比较,各药物处理组与阴性对照组比较,各药物处理组组间比较,细胞存活率均差异显著(P<0.05);各组内用药48 h、72 h分别与作用24 h比较,各组内作用72 h与作用48 h比较,细胞存活率差异显著(P<0.05);细胞G_0/G_1期和G_2/M期分布,阴性对照组与空白对照组比较,差异显著(P<0.05),不同浓度肉苁蓉正丁醇萃取物处理组(1 600μg/mL、800μg/mL、400μg/mL)分别与阴性对照组比较,差异均显著(P<0.05)。结论:肉苁蓉正丁醇萃取物可在短时间内对Dox造HKC细胞凋亡的毒性有明显抑制作用,且具有浓度依赖性,其作用机制可能是通过抑制Dox阻断细胞进一步分裂进入下一个细胞周期,促进HKC细胞增殖。
Objective:To study of N-butanol extracts in different concentration from Herba Cistanches on HKC cell injuryinduced by doxorubicin(Dox). Methods:N-butanol extracts in different concentrations of Herba Cistanches were used to the HKC injury cell model induced by Dox at different times in vitro,respectively,and blank and negative control groups were set. Cell survival rate was measured by MTT. And cell cycle distribution was detected by flow cytometry with single straining of pyridine iodide. Results:The negative control group was compared with the blank control group,the treatment groups were compared with the negative control group,and the treatment group was compared with the control group,there were statistical significant differences(P <0.05) in cell survival rate. In the same treatment group,compared with that after the treatment for 24 h,the survival rate of cells were significantly different(P <0.05) after the treatment for 48 h and 72 h,respectively. In the same treatment group,compared with that after the treatment for 48 h,the survival rate of cells were significantly different(P<0.05)after the treatment for 72 h. There was statistical difference(P<0.05)between negative control group and blank control group in cell distribution of G0/G1 and G1/G2. The groups of N-butanol extracts with concentrations of1 600 μg/mL,800 μg/mL,400 μg/mL had statistical difference(P<0.05) in cell distribution of G0/G1 and G1/G2 compared with negative control group. Conclusion:N-butanol extracts of Herba Cistanches could significantly inhibit apoptosis toxicity in HKC cell induced by Dox in a short period time,and showed significant concentration dependence. The possible mechanism was to inhibit the further division into the next cell cycle and promote the HKC cell proliferation by inhibiting Dox.
