麦门冬汤对特发性肺间质纤维化小鼠肺组织PPARγ表达和可溶性胶原蛋白含量的影响 被引量:4

Effects of Maimendong Decoction(麦门冬汤) on PPARγ Expression and Soluble Collagen Content in Lung Tissue of Mice with Idiopathic Pulmonary Interstitial Fibrosis

摘要 目的探讨麦门冬汤治疗特发性肺间质纤维化的可能作用机制。方法BABL/c小鼠随机分为模型组、实验组和对照组,每组15只。模型组和实验组小鼠分别在气管内连续5次滴注博来霉素40 mg/kg,每2天1次,建构特发性肺间质纤维化小鼠模型。实验组滴注博来霉素48 h后采用麦门冬汤(浓度为0.6 g/ml)混悬液按照2 ml/200 g灌胃,模型组和对照组以等容积生理盐水灌胃,均每日1次,灌胃2周。干预后检测各组小鼠肺功能[包括吸气峰流速(PIF)、呼气峰流量(PEF)及每分钟通气量(MV)]、肺组织可溶性胶原蛋白含量及过氧化物酶体增殖物激活受体γ(PPARγ)mRNA及蛋白表达水平。结果与正常组比较,模型组小鼠PIF、PEF及MV均显著下降,肺组织中PPARγmRNA及蛋白表达水平均降低,可溶性胶原蛋白显著升高(P<0.05或P<0.01);与模型组比较,实验组小鼠PIF、PEF及MV均显著升高,肺组织中PPARγmRNA及蛋白表达水平升高,可溶性蛋白显著降低(P<0.05)。结论麦门冬汤可能通过升高肺组织中PPARγ表达,降低可溶性胶原蛋白含量,从而改善特发性肺间质纤维化小鼠肺功能。 Objective To investigate the possible mechanism of Maimendong Decoction (麦门冬汤) in the treatment of idiopathic pulmonary interstitial fibrosis. Methods BABL /c mice were randomly divided into a model group,an experimental group and a control group,with 15 mice in each group. In the model group and the experimental group,bleomycin 40 mg /kg was instilled in the trachea for 5 times every 2 days to construct a mouse model of idiopathic pulmonary interstitial fibrosis. In the experimental group,after 48 hours of instilling bleomycin,the suspension of Maimendong Decoction ( concentration of 0. 6 g /ml) was administered 2 ml /200 g. The model group and the control group were intragastrically administered with equal volume of normal saline,once a day,gavage for 2 weeks. Pulmonary functions including peak inspiratory flow ( PIF),peak expiratory flow ( PEF) and minute ventilation ( MV),soluble collagen content in lung tissue and peroxisome proliferator activated receptor γ( PPARγ) mRNA and protein expression levels of each group were measured after intervention. Results Compared with the normal group,the PIF,PEF and MV of the model group were significantly decreased,the expression of PPARγ mRNA and protein in the lung tissue decreased,and the soluble collagen was significantly increased ( P < 0. 05 or P < 0. 01). Compared with the model group,PIF,PEF and MV were significantly increased in the experimental group. PPARγ mRNA and protein expression levels were increased in lung tissue,and soluble protein was significantly decreased ( P < 0. 05). Conclusion Maimendong Decoction may improve the lung function of mice with idiopathic pulmonary fibrosis by increasing the expression of PPARγ in lung tissue and decreasing the content of soluble collagen.
