摘要 目的对色素上皮衍生因子(PEDF)在急性脊髓损伤中的作用及其机制进行初步探讨。方法建立脊髓损伤大鼠模型,通过BBB评分法评估大鼠的神经运动功能,尼氏染色法观察脊髓的组织学变化,免疫荧光法和免疫印迹法观察Fas/FasL的表达。结果治疗后7、14 d,SCI + PEDF组的BBB评分较SCI + NS组显著升高,差异有统计学意义(P<0.05)。尼氏染色结果显示,SCI + PEDF组的空洞率较SCI+NS组明显减少(P<0.05)。免疫印迹结果显示,治疗后3 d SCI + PEDF组Fas/FasL的蛋白表达呈降低趋势,SCI + NS组Fas/FasL的蛋白表达呈持续升高趋势。免疫荧光结果显示,治疗后3 d SCI + PEDF组中的Fas/FasL表达较SCI + NS组明显减少,差异有统计学意义(P<0.05)。结论PEDF能抑制大鼠脊髓后的脊髓神经细胞凋亡,其作用机制可能与Fas/FasL信号通路有关。 Objective To explore the role of pigmant epithelinm derived facter ( PEDF) in spinal cord injury ( SCI) and its mechanism. Methods A SCI rat model was established and assessed for neuromotor function in rats by Basso, Beattie and Bresnahan (BBB) scores. Nissl staining was used to observe histological changes in the spinal cord. The ex pression of Fas/FasL was observed by immunofluorescence and protein electrophoresis. Results Compared with the SCI + NS group, the SCI + PEDF group had a significantly higher BBB score 7 and 14 days after treatment (P <0.05). In Nissl staining, the void rate of the SCI + PEDF group was significantly lower than that of the SCI + NS group (P <0.05). The expression of Fas/FasL decreased in the SCI + PEDF group after 3 days, and the level of Fas/FasL in the SCI + NS group continued to increase. Immunofluorescence showed that the expression of Fas/FasL in SCI + PEDF group was sig nificantly lower than that in SCI + NS group after 3 days ( P < 0. 05 ). Conclusions PEDF can inhibit the apoptosis of spinal cord neurons in rat spinal cord, which may be related to Fas/FasL signaling pathway.
