油茶根际高效解磷细菌NC285液体发酵培养条件的优化 被引量:6

Optimization of Liquid Fermentation Conditions for Efficient Phosphate Solubilizing Bacteria NC285 in Camellia oleifera Rhizosphere

摘要 磷是植物生长的必需元素之一,土壤中绝大多数的磷都以植物难以吸收的形态存在,导致了植物无法正常生长,而解磷细菌具有溶解难溶性磷的特性,因此,开发高效解磷细菌对提高磷的利用率具有重要的意义。为了探究油茶(Camellia oleifera Abel.)根际解磷细菌的最适生长条件,采用单因素试验分别考察了碳源、氮源、温度、pH、接种量以及溶氧量对该菌株生长的影响,并在单因素的基础上,采用正交试验对其溶磷条件进行优化。结果表明该菌株的最适碳源为葡萄糖,最适氮源为(NH4)2SO4,最佳培养条件分别为温度28℃,初始pH值7.5,接种量1%,溶氧量100 mL。解磷菌在优化后的培养基中的溶磷能力明显提高,培养基有效磷含量高达586.73 mg/L,比优化前提高了45.16%。 Phosphorus is one of the essential elements for plant growth.But most of the phosphorus in the soil exists in a form that is difficult for plants to absorb,which results in the fact that plants can not grow normally.Phosphate-dissolving bacteria have the property of dissolving insoluble phosphorus.Therefore,the development of high-efficiency phosphate-dissolving bacteria is of great significance for improving the utilization of phosphorus.To clear out the optimum growth conditions of the phosphorus-dissolving bacteria in the rhizosphere of Camellia oleifera Abel.,the effects of optimal carbon source,nitrogen source,temperature,pH,inoculation quantity and aeration quantity on the growth of the strain were investigated by single factor test.The optimum conditions for phosphorus-solubilizing bacteria strain NC285 were optimized by orthogonal experiment on the basis of single factor test.The results showed that the optimum culture conditions were as follows:glucose used as carbon source and(NH4)2SO4 as nitrogen source,the initial pH 7.5,inoculation quantity 1% and the culture temperature 28 ℃ for the strain NC285.The phosphate solubilizing ability of NC285 in the optimized medium increased significantly.After optimized,the effective phosphorus content of medium reached 586.73 mg/L,which was 45.16% higher than that before optimization.
