中国美味蘑菇石油醚提取物体外抗肿瘤活性研究 被引量:2

摘要 采用细胞划痕方法检测中国美味蘑菇石油醚提取物对A549细胞体外迁移能力的影响,根据克隆形成的数量评估石油醚提取物对A549细胞的增殖能力的影响;并测定A549细胞增殖效率、细胞凋亡率以及BCL-2、Bcl-xl、Cyto-c、Caspase-3和AIFM1基因的表达水平。结果表明:中国美味蘑菇石油醚提取物可降低非小细胞肺癌A549细胞的迁移能力,且呈剂量依赖性;石油醚提取物能够抑制非小细胞肺癌A549细胞的增殖,同样呈剂量依赖性;石油醚提取物处理后的非小细胞肺癌A549细胞凋亡率明显增加;BCL-2和Bcl-xl基因表达水平降低,Cyto-c、Caspase-3和AIFM1表达水平提高。因此,石油醚提取物可以有效抑制非小细胞肺癌A549细胞的增殖,降低细胞迁移能力和克隆形成,诱导细胞凋亡。 The effects of petroleum ether extracts from the fruiting bodies of Agaricus sinodeliciosus were studied with non-small cell lung cancer A549 cells on cell migration,cloning,proliferation and apoptosis.Scratch wound assay was used to detect the migration activity of A549 cells.After following treatment,the proliferation ability of A549 cells with petroleum ether extract was further evaluated through the colony formation assay,and then the proliferation inhibition rate was examined by alamar blue assay.Measurement of apoptosis was examined by annexinV-FITC PI staining.Meanwhile,the mRNA levels of BCL-2,Bcl-xl,Cyto-c and Caspase-3 were examined by qRT-PCR.The results showed that the migration ability of non-small cell lung cancer A549 cells in a dose-dependent manner reduced greatly when they had been treated with the petroleum ether extract from the fruiting bodies of A.sinodeliciosus.The petroleum ether extract inhibited the proliferation of non-small cell lung cancer A549 cells in a dose-dependent manner.After treated with petroleum ether extract,the numbers of apoptotic A549 cells were significantly increased.Further studies showed that the gene levels of BCL-2 and Bcl-xl were decreased,while the gene levels of Cyto-c,Caspase-3 and AIFM1 were increased.Therefore,the results suggested that the petroleum ether extract might be one potential drug in non-small cell lung cancer treatment by inhibiting proliferation,blocking cell migration ability and colony formation,inducing apoptosis.
