摘要
目的观察婴儿血管瘤(Infantile hemangioma,IH)组织中微小RNA(micro RNA,miR)-130a、B淋巴细胞瘤-2(Bcl-2)的表达变化。方法 33例IH患者,其中增殖期17例、退化期16例,留取IH组织、瘤旁正常皮肤组织。采用荧光实时定量PCR法检测IH组织、瘤旁正常皮肤组织中miR-130a和Bcl-2 mRNA,采用SP免疫组织化学法检测IH组织、瘤旁正常皮肤组织BCl-2蛋白,分析IH组织miRNA-130a、Bcl-2 mRNA表达的相关性。结果 IH组织、瘤旁正常皮肤组织中miR-130a相对表达量分别为3.71±1.23、 0.87±0.21,退化期、增殖期IH组织miRNA-130a相对表达量分别为3.68±1.40 、3.72±1.63;IH组织及瘤旁正常皮肤组织Bcl-2 mRNA相对表达量分别为1.213±0.272 、0.614±0.22,退化期、增殖期IH组织Bcl-2 mRNA相对表达量比较分别为3.68±1.40 、 3.72 ±1.63,与瘤旁正常皮肤组织比较,退化期、增殖期IH组织miRNA-130a及Bcl-2 mRNA相对表达量均升高( P 均<0.05)。与瘤旁正常皮肤组织比较,IH组织Bcl-2蛋白光密度值和阳性面积率均升高( P 均<0.05);与退化期IH组织、瘤旁正常皮肤组织比较,增殖期IH组织Bcl-2蛋白光密度值和阳性面积率均升高( P 均<0.05)。IH组织中miR-130a 与Bcl-2 mRNA的表达呈正相关( r =0.451, P =0.008)。结论IH组织miR-130a、Bcl-2 mRNA及蛋白表达均升高,增殖期IH组织Bcl-2蛋白表达高于退化期。miR-130a、Bcl-2可能协同促进IH的发生、发展。
Objective To observe the expression of microRNA-130a (miR-130a )and B lymphocytoma-2 (Bcl-2) in infantile hemangioma (IH). Methods Thirty-one cases of patients with IH were enrolled, including 17 cases in proliferative stage and 16 cases in degenerative stage. The expression of miR-130a and Bcl-2 in IH tissues and adjacent normal skin tissues were detected by real-time quantitative fluorescence PCR (qRT-PCR). Bcl-2 protein in IH tissues and adjacent normal skin tissues was detected by SP immunohistochemistry. The correlation between the expression of miR-130a and Bcl-2 in IH tissues were analyzed. Results The relative expression levels of miR-130a in IH tissues and adjacent normal skin tissues were 3.71±1.23 and 0.87±0.21, respectively. The relative expression levels of miRNA-130a in the degenerative and proliferative IH tissues were 3.68±1.40 and 3.72±1.63, respectively. The relative expression levels of Bcl-2 mRNA in the IH tissues and adjacent normal skin tissues were 1.213±0.272 and 0.614±0.22, respectively. The relative expression levels of Bcl-2 mRNA in the degenerative and proliferative IH tissues were 3.68±1.40 and 3.72±1.63, respectively. The relative expression levels of miR-130a and Bcl-2 mRNA in the IH tissues were significantly higher than those in adjacent normal skin tissues (all P <0.05).Bcl-2 protein optical density and positive area in the IH tissues were higher than those in the adjacent normal skin tissues (all P <0.05);Bcl-2 protein optical density and positive area in IH tissues during proliferation were higher than those in degenerative IH tissues and adjacent normal skin tissues (all P <0.05). There was a positive correlation between the expression of miR-130a and Bcl-2 mRNA in IH tissues ( r =0.451, P = 0.008 ). Conclusions The expression levels of and Bcl-2 in IH tissues increase, and the expression of Bcl-2 in IH tissues during proliferation was higher than that in degeneration. miR-130a and Bcl-2 may synergistically promote the occurrence and development of IH.
