卵巢癌对转移前网膜微血管内皮的活化作用

摘要 目的 探讨卵巢癌对转移前网膜微血管内皮细胞的活化作用。方法免疫组化法检测37例原发性卵巢癌患者网膜组织的CD34和CD105表达,评估微血管密度,并分析其与患者临床病理特征的相关性。蛋白印迹法检测卵巢癌细胞培养上清(条件培养液)对原代网膜内皮细胞CD105表达水平的影响,未荷瘤裸鼠腹腔注射卵巢癌细胞培养上清后免疫组化法检测网膜CD31表达,分析卵巢癌对转移前网膜微血管密度的影响,Transwell实验检测卵巢癌细胞培养上清对微血管内皮细胞侵袭能力的影响。结果卵巢癌网膜转移患者网膜微血管密度较无网膜转移患者显著增加,卵巢癌细胞培养上清可以增加内皮细胞CD105的表达,促进小鼠网膜血管新生,并增强内皮细胞侵袭能力。结论卵巢癌可以促进转移前网膜微血管内皮活化形成利于转移灶形成的微环境。 Objective To investigate the effects of ovarian cancer on pre-metastatic omental microvascular endothelial cells. Methods CD34 and CD105 were detected by immunohistochemical analyses to evaluate the microvessel density in 37 omental specimens obtained from patients with primary ovarian cancer and the correlation between the microvessel density and the clinicopathological characteristics of ovarian cancer patients was analyzed.Next,the effect of cancer cells on the expression of CD105 in primary endothelial cells derived from human omentum was detected by Western blotting.Then animal experiments were used to analyze the effect of ovarian cancer cell supernatants(conditioned medium)on the omental microvessel density.Finally,the effect of cancer cells on the invasion ability of microvascular endothelial cells was detected by transwell assays. Results The microvessel density in human omental specimens was significantly increased in metastatic omentum.The supernatants of ovarian cancer cells increased the expression level of CD105 in the primary endothelial cells,enhanced the invasion of endothelial cells,and promoted omental angiogenesis in vivo . Conclusion Ovarian cancer can activate the omental microvascular endothelial cells even before metastatic colonization,which may facilitate the subsequent metastasis.
