摘要
目的探讨丹参酮ⅡA在体外对人膀胱癌J82细胞增殖、凋亡、侵袭及迁移的影响。方法以不同浓度的丹参酮ⅡA处理J82细胞,分别采用细胞计数试剂盒(CCK-8)法、膜联蛋白V-异硫氰酸荧光素(Annexin V-FITC)/碘化丙锭(PI)双标记流式检测法、Transwell侵袭实验及划痕实验检测J82细胞的增殖、凋亡、侵袭及迁移情况。结果丹参酮ⅡA可抑制J82细胞增殖,且呈浓度与时间依赖性(均P<0.05)。1、3、5μmol/L丹参酮ⅡA处理组总凋亡率分别为12.23%、14.74%、18.40%,均高于对照组的3.36%(P<0.05),且呈浓度依赖性(P<0.05)。对照组、1μmol/L组、3μmol/L组、5μmol/L组细胞侵袭数比较,差异有统计学意义(P<0.05),且呈浓度与时间依赖性(均P<0.05)。对照组、1μmol/L组、3μmol/L组、5μmol/L组划痕后12、24、36h划痕愈合率比较,差异均有统计学意义(均P<0.05),呈浓度依赖性(P<0.05)。结论丹参酮ⅡA在体外可抑制膀胱癌J82细胞增殖,促进其凋亡,降低其侵袭和迁移能力。
Objective To investigate the effects of tanshinoneⅡA on the proliferation,apoptosis,invasion and migration of human bladder carcinoma J82 cells in vitro.Methods The proliferation,apoptosis,invasion and migration of J82 cells treated with tanshinoneⅡA of various doses were evaluated by CCK-8 assay,flow cytometric analysis with Annexin V-FITC/PI staining,Transwell invasion assay and wound healing assay,respectively.Results The proliferation of J82 cells was inhibited by tanshinoneⅡA in a dose-and time-dependent manner(P<0.05).The apoptosis rates of J82 cells in 1μmol/L group(12.23%),3μmol/L group(14.74%)and 5μmol/L group(18.40%)were all higher than that in control group(3.36%,P<0.05),and the apoptosis rate increased with the increase of drug dosage(P<0.05).The numbers of invasive cells among control group,1μmol/L group,3μmol/L group and 5μmol/L group were significantly different(P<0.05),and the number of cells decreased with the increase of drug dosage and treatment time(P<0.05).The wound closure rates were significantly different among control group,1μmol/L group,3μmol/L group and 5μmol/L group at the 12 h,24 h and 36 h after scratch(P<0.05),and the wound closure rate decreased with the increase of drug dosage(P<0.05).Conclusion TanshinoneⅡA can significantly inhibit proliferation,invasion,migration and induce apoptosis of human bladder carcinoma J82 cells in vitro.
