吸入性麻醉药对宫颈鳞癌细胞生物学行为的影响

摘要 目的探讨吸入性麻醉药对宫颈鳞癌细胞生物学行为(细胞增殖、迁移和侵袭等)的影响。方法将宫颈鳞癌Caski细胞暴露于不同剂量(1%、2%、3%)的吸入性麻醉药(异氟醚或七氟醚),采用细胞计数试剂盒-8(CCK-8)法、免疫荧光法分别检测细胞增殖能力和Ki-67表达,Transwell小室法检测细胞侵袭及迁移能力。结果与对照组(仅混合气体)比较,七氟醚对Caski细胞增殖及Ki-67表达均无影响(均P>0.05);1%、2%异氟醚能促进Caski细胞的增殖(均P<0.05),其中2%异氟醚组Caski细胞中Ki-67表达明显增加(P<0.05)。与对照组比较,2%、3%七氟醚或异氟醚均能促进细胞侵袭和迁移能力(均P<0.05);1%七氟醚或异氟醚对细胞侵袭和迁移能力均无影响(均P>0.05)。结论吸入性麻醉药(七氟醚或异氟醚)能促进宫颈鳞癌细胞的侵袭和迁移;同时异氟醚能促进宫颈鳞癌细胞的增殖能力。 Objective To investigate the effects of inhalational anesthetics on proliferation,migration and invasion of squamous cervical cancer cells.Methods Squamous cervical cancer Caski cells were exposed to 1%,2%or 3%sevoflurane and isoflurane,respectively.Cell proliferation and Ki-67 expression were detected by CCK-8 assay and immunofluorescence assay,respectively.Cell invasion and migration were detected by Transwell assay.Results Compared with control group,1%and 2%isoflurane significantly enhanced the proliferation of Caski cells(P<0.05),2%isoflurane also increased the expression of Ki-67 in Caski cells(P<0.05);but there was no effect of sevoflurane on the proliferation and Ki-67 expression of Caski cells(P>0.05).After 2%and 3%sevoflurane or isoflurane treatment,cell migration and invasion ability were promoted,compared to the control group(P<0.05);however,1%sevoflurane or isoflurane had no effect on the migration and invasion of Caski(P>0.05).Conclusion Inhalational anesthetics sevoflurane and isoflurane can enhance migration and invasion of squamous cervical cancer Caski cells.Isoflurane also can promote the proliferation of squamous cervical cancer cells.
