METTL3在结直肠癌组织中的表达及其临床意义

Overexpression of METTL3 predicts poor prognosis in colorectal cancer

目的探讨甲基转移酶样3(METTL3)在结直肠癌组织中的表达及临床意义。方法收集67例病理检查诊断为结直肠癌患者的结直肠癌组织、配对癌旁正常组织及肝脏、淋巴结转移组织,采用免疫组化技术,检测METTL3蛋白的表达情况,RT-PCR技术检测METTL3 mRNA相对表达量情况。通过ROC曲线分析确定METTL3 mRNA相对表达量的截断值,以此分析METTL3 mRNA的表达与各临床病理因素之间的关系,并分析METTL3 mRNA的表达水平与结直肠癌患者生存预后的关系。结果相比于正常组织,METTL3 mRNA在结直肠癌组织中表达明显升高(P<0.05),且METTL3 mRNA相对表达量的截断值为2.85。免疫组化结果显示,与正常结直肠组织相比,METTL3蛋白在结直肠癌组织中核染明显。METTL3 mRNA的高表达(相对表达量>2.85)与淋巴结转移密切相关(P<0.01),与性别、年龄、肿瘤大小、组织分化、远处转移、侵袭深度无相关(均P>0.05)。Kaplan-Meier生存分析结果显示,与METTL3 mRNA低表达的结直肠癌患者比较,METTL3 mRNA高表达患者在随访的4年间生存率明显降低,差异有统计学意义(P<0.01)。结论METTL3在结直肠癌组织中呈高表达,与患者淋巴结转移及生存预后有关。

Objective To explore the expression of METTL3 in colorectal cancer(CRC)and its clinical implication in colorectal cancer.Methods Immunohistological staining(IHC)and quantitative real-time polymerase chain reaction(qRT-PCR)were performed to detect METTL3 expression in tumor tissue samples from 67 CRC patients admitted in Kanghua Hospital from 2013 to 2018.The correlation between METTL3 expression and clinicopathological features of CRC and the overall survival(OS)of patients was examined by univariate and multivariate analyses.Results The expression of METTL3 was significantly associated with lymph node metastasis(P<0.01),but not correlated with age,gender,tumor diameter,depth of invasion,histological differentiation and distant metastasis.The median OS for CRC patients with high or low METTL3 expression were 23 months and 43 months(P<0.01).Conclusion The results suggest that METTL3 may contribute to clinical tumor progression and it might be used as a prognostic biomarker in colorectal cancer.
