摘要
目的:探讨掌叶半夏有效提取物对宫颈癌大鼠癌组织中Survivin、Caspase-3、及Caspase-7的影响。方法:选取Wistar大鼠(雌性)50只,随机选10只为对照组,其余40只建宫颈癌模型,建模成功大鼠(n=36)随机分为模型组、阳性对照组、观察组各12例。模型组未进行任何治疗,阳性对照组行顺铂2mg/kg腹腔注射,观察组用掌叶半夏有效提取物40mg/kg灌胃,均1次/d,8周后处死大鼠,取宫颈癌组织体积进行测量,并对癌组织中Survivin、Caspase-3及Caspase-7的表达进行检测。结果:与模型组比较,观察组及阳性对照组体积均明显减小(P<0.01);观察组体积与阳性对照组比较,差异无统计学意义(P>0.05);观察组与模型组比较,Survivin蛋白表达明显降低,Caspase-3及Caspase-7蛋白表达明显升高(P<0.05);观察组Survivin、Caspase-3及Caspase-7蛋白表达与阳性对照组比较(P>0.05);癌病灶体积与Survivin蛋白表达呈正相关,与Caspase-3及Caspase-7蛋白表达呈负相关。结论:掌叶半夏有效提取物能使宫颈癌大鼠病灶的体积缩小,其作用机制之一可能是能够降低癌组织Survivin表达、升高Caspase-3、Caspase-7表达等细胞凋亡蛋白所致。
Objective:To investigate the effect of effective extract of Pinellia ternata on Survivin,Caspase-3 and Caspase-7 in cervical carcinoma rats.Methods:50 wistar rats(female)were selected.Ten rats were randomly 10 as control group,the remaining 40 were all building models of cervical cancer.Thirty-six rats were randomly divided into three groups:the model group,the positive control group and the observation group(n=12).No treatment was given in the model group,the positive control group was intraperitoneally injected with cisplatin 2 mg/kg,in the observation group,the effective extract of Pinellia ternata,40 mg/kg,was administrated by stomach,each one per day.The rats were killed after 8 weeks.The tissue volume of cervical cancer was measured,the expression of survivin caspase-3 and caspase-7 were detected.Results:Compared with model group,the volume of observation group and positive control group was significantly decreased(P<0.01).The volume of the observation group was compared with that of the positive control group,the difference was not statistically significant(P>0.05).The observation group was compared with the model group,the expression of survivin protein was significantly decreased while the expression of Caspase-3 and Caspase-7 protein were significantly increased(P<0.05).The expression of survivin,Caspase-3 and Caspase-7 protein in the observation group was compared with those in the positive control group(P>0.05).The tumor volume was positively correlated with the expression of survivin protein,and negatively correlated with the expression of Caspase-3 and Caspase-7 protein.Conclusion:The effective extract of Pinellia ternata can reduce the volume of cervical cancer in rats.One of its mechanisms may be that it can decrease the expression of survivin and increase the expression of Caspase-3 and Caspase-7.
