摘要
目的:观察肠愈灌肠方对溃疡性结肠炎大鼠细胞凋亡因子Bcl-2、Bax蛋白表达,揭示肠愈灌肠方治疗溃疡性结肠炎的作用机制。方法:将60只SPF大鼠按随机数字表法分为空白组、模型组、美沙拉嗪组、肠愈灌肠方组,每组15只。采用2,4,6-三硝基苯磺酸(TNBS)复制溃疡性结肠炎大鼠动物模型,予肠愈灌肠方干预,并与西药美沙拉嗪对比。观察治疗过程中各组大鼠DAI评分,采用Western-Blot法检测大鼠Bcl-2、Bax蛋白表达水平。结果:与模型组比较,肠愈灌肠方和美沙拉嗪组能明显降低溃疡性结肠炎大鼠DAI评分(P<0.05);与空白组比较,模型组大鼠Bcl-2减少、Bax增多(P<0.05);与模型组比较,肠愈灌肠方组、美沙拉嗪组大鼠Bcl-2表达水平均升高,Bax减弱(P<0.05);与美沙拉嗪组比较,肠愈灌肠方组大鼠Bcl-2表达增多,Bax减弱,但差异无统计学意义(P>0.05)。结论:肠愈灌肠方治疗溃疡性结肠炎疗效确切,其作用机制可能与调节Bcl-2、Bax蛋白因子表达有关。
Objective:To observe the expression of apoptotic factors Bcl-2 and Bax in rats with ulcerative colitis treated by Changyu Guanchang Recipe,and to reveal the mechanism of Changyu Guanchang Recipe in treating ulcerative colitis.Methods:60 SPF rats were randomly divided into blank group,model group,Mesalazine group and Changyu Guanchang Recipe group with 15 rats in each group.The rat model of ulcerative colitis was induced by 2,4,6-trinitrobenzene sulfonic acid(TNBS)and intervened by Changyu Guanchang Recipe,which was compared with Mesalazine.The DAI scores of rats in each group were observed during the treatment.The expression levels in Bcl-2 and Bax protein of rats were detected with Western-Blot method.Result:Compared with model group,Changyu Guanchang Recipe group and Mesalazine group could significantly reduce the DAI score of ulcerative colitis rats(P<0.05);compared with blank group,Bcl-2 decreased and Bax increase of model group(P<0.05);compared with model group,Changyu Guanchang Recipe group and Mesalazine group,Bcl-2 was increased.Compareing the Mesalazine group,the expression of Bcl-2 increased and Bax decreased in the Changyu Guanchang Recipe group,but there was no significant difference(P>0.05).Conclusion:Changyu Guanchang Recipe is effective in treating ulcerative colitis,and its mechanism may be related to regulating the expression of Bcl-2 and Bax protein.
