油脂中β-胡萝卜素含量的检测

The Determination of β-Carotene in Oil by High Performance Liquid Chromatography

建立固相萃取-超高效液相色谱法测定油脂中β-胡萝卜素的方法。样品经正己烷溶解,经中性氧化铝柱层析净化、浓缩、二氯甲烷定容。以BEHC18色谱柱分析,甲醇︰乙腈(v/v90︰10)为流动相,在0.3mL/min的流速下,选择波长448nm检测。β胡萝卜素的加标回收率为91.2%~98.2%,变异系数为1.31%~2.66%,线性范围为1.0~100μg/mL,相关系数为0.9999,方法的检出限为1.0mg/kg。该方法检测流程简便,净化效果好,灵敏度高,可满足实际样品分析的需求。

An analytical method was developed for the determination ofβ-Carotene in oil.Sample was dissolved by hexane,purified by aluminium oxide column.After concentration,then volumed with methylene chloride.The mobile phase was methanol:acetonitrile(v/v 90︰10).Sample solution was separated by BEHC 18 column.The flow rate of mobile phase was 0.3 mL/min.The Analyte was measured by 448 nm.The range of recoveries was 91.2%~98.2%.The relative standard deviation was1.31%~2.66%.The range of calibration curve was 1.0~100μg/mL.The correlation coefficient of calibration curve was 0.9999.The limitation of determination was 1.0 mg/kg.This method simplified the experimental procedure,modified the result of purification and also increase the sensitivity of the determination.
