摘要
目的探究细胞外信号调节激酶5(ERK5)对人体外血小板聚集的影响和体内血栓形成的影响。方法将本院获得的人血50份,制备血小板悬浮液,随机将血小板悬浮液分为两组,对照组和干预组,每组25份。干预组血小板悬浮液给予ERK5,对照组血小板悬浮液给予等量的生理盐水,然后采用血小板聚集仪检测体外血小板的凝集情况。购买SPF级雌性BALB/c型小鼠40只,建立FeCl3颈动脉血栓小鼠模型,将FeCl3颈动脉血栓小鼠模型随机分为两组,实验组和空白组,每组20只,实验组静脉给予ERK5,对照组静脉给予特异性抑制剂XMD8-92,观察小鼠颈动脉血栓形成时间,并比较两组是否有差异。结果 50份血小板悬浮液,对照组血小板凝集5份,干预组血小板凝集23分。两组差异有统计学意义(P<0.05)。40例FeCl3颈动脉血栓小鼠模型中给予ERK5特异性抑制剂XMD8-92的实验组小鼠第一次血栓形成平均时间为(12.56±0.47)min;给予DMSO溶液的空白组第一次血栓形成平均时间为(7.62±0.89)min,两组差异有统计学意义(P<0.05)。结论 ERK5在体外和体内血小板的凝集过程中发挥重要作用。
Objective To investigate the effect of extracellular signal regulation kinase 5(ERK5)on platelet aggregation in vitro and the effect of thrombosis in vivo.Methods 50 human blood was obtained from the hospital,the platelet suspension was prepared,and the platelet suspension was randomly divided into two groups,the control group and the intervention group,25 in each group.The platelet suspension of the intervention group was given ERK5,the same amount of saline was given to the platelet suspension in the control group,and platelet aggregation was used to detect the agglutination of platelets in vitro.40 of SPF female BALB/c mice were purchased,a mouse model of FeCl3 carotid artery thrombosis was established,and the model of FeCl3 carotid thrombosis mice was randomly divided into two groups,the experimental group and the Blank group,20 in each group,the veins of the experimental group were given ERK5,and the control group intravenous was given a specific inhibitor XMD8-92,The time of carotid artery thrombosis in mice was observed,and whether there were differences between the two groups.Results 50 platelet suspensions,5 platelet agglutination in the control group and 23 points of platelet agglutination in the intervention group.The differences between the two groups were statistically significant(P<0.05).In 40 cases of FeCl3 carotid thrombosis mice,the average time of first thrombosis in mice with ERK5 specific inhibitor XMD8-92 was(12.56±0.47)min,and the average time of first thrombosis in the blank group given DMSO solution was(7.62±0.89)min,The differences between the two groups were statistically significant(P<0.05).Conclusion ERK5 plays an important role in the agglutination process of platelets in vitro and in vivo.
