摘要
目的:研究乌兰温都苏-11丸对自发性高血压大鼠(SHR)心、肾、血管组织中AT1R、AT2R蛋白表达的影响。方法:SHR大鼠60只和同源相匹配的Wistar Kyoto(WKY)大鼠10只,随机分为模型组、对照组、缬沙坦胶囊组、丹参酮Ⅱ_A硫酸钠注射液组、乌兰温都苏-11丸低、中、高剂量组,每组10只。对照组和模型组给予蒸馏水,缬沙坦胶囊组、蒙药乌兰温都苏-11丸低、中、高剂量组每天同时按相应剂量灌胃药物、丹参酮Ⅱ_A磺酸钠注射液组相应剂量腹腔注射药物。给药治疗4周后,采用蛋白质免疫印迹方法分别检测大鼠心、肾、血管中血管紧张素Ⅱ受体AT1R、AT2R蛋白表达水平。结果:Western blot的检测结果表明乌兰温都苏-11丸干预显著降低SHR大鼠心、肾、血管的血管紧张素Ⅱ受体AT1R蛋白表达水平,同时显著增加血管紧张素Ⅱ受体AT2R蛋白表达水平。结论:乌兰温都苏-11丸治疗能够明显抑制AT1R蛋白表达,并显著激活AT2R蛋白表达。其作用机制可能是可通过拮抗血管紧张素Ⅱ受体AT1R的同时激活血管紧张素Ⅱ受体AT2R调节高血压,对高血压引起的靶器官损伤有一定的防治作用。
objective: To study the effects of Wulan Wendusu-11 pill on the expression of AT1R and AT2R proteins in heart, kidney and blood vessels of SHR rats. Methods: Sixty SHR rats and 10 Wistar Kyoto (WKY) rats that matched with each other were randomly divided into model group,control group, valsartan capsule group,tanshinone II Asodium sulfate injection group, Wulan WenduSu-11 pill low,middle and high dose groups, W in each group. The control group and model group were given distilled water,valsartan capsule group, mongolian medicine Wulan Wendusu-11 pill low, middle, and high dose groups were given corresponding doses of gavage drugs daily, and corresponding doses of tanshinone U A sodium injection group were given intraperitoneally. After 4 weeks, the expression levels of angiotensin U receptor ATI R and AT2R in rat heart, kidney and blood vessels were detected by Western blot. Results: Significant downregulation of angiotensin U type 1 receptor protein expression in SHR rats and up-regulation of angiotensin U type 2 receptor protein expression. Conclusion: Treatment with Wulan Wendusu-11 pill can significantly inhibit ATI R protein expression and significantly activate AT2R protein expression. The mechanism of action may be to exert an antihypertensive effect by antagonizing the angiotensin U receptor ATIR while activating the angiotensin U receptor AT2R,It has certain preventive and therapeutic effects on target organ damage caused by hypertension.
