摘要
目的:建立妇科洗剂中总生物碱含量的测定方法。方法:采用酸性染料比色法测定妇科洗剂总生物碱含量,并用正交实验筛选最优反应条件。结果:酸性染料比色法测定总生物碱含量的最佳条件为0.05%溴酚蓝染料用量1.5 mL,pH4.0的柠檬酸-柠檬酸钠缓冲溶液用量7.5 mL,氯仿萃取2次,每次5 mL,检测波长为405 nm。在浓度范围为0.022 4~0.112 0 g·L^-1内盐酸小檗碱对照品的吸光度与浓度线性关系良好。结论:此法灵敏度较好,准确度高,操作简便,能为妇科洗剂质量控制提供依据。
objective:To establish a method for determination of total alkaloid content in gynecological lotion.Methods:The total alkaloid content in gynecological lotion was determined by acid dye colorimetry,and the optimal reaction conditions were screened by orthogonal experiment. Results:The optimal conditions of determinating total alkaloid content by acid dye colorimetry were as following,the amount of 0.05% bromophenol(as blue dye)was1.5 mL;the amount of pH4.0 citric acid-sodium citrate(as buffer solution)was 7.5 mL;chloroform extraction was extracted twice,5 mL each time;the detection wavelength was 405 nm.The absorbance and concentration of berberine hydrochloride(as reference substance)in concentration range of 0.022 4-0.112 0 g·L^-1 showed a good linear relationship. Conclusion:This method has good sensitivity,high accuracy and simple operation,which can provide basis for quality control of gynecological lotion.
