摘要
目的:观察丹连消痤散外用对痤疮模型兔耳NF-κB(核转录因子kappa B)、p38MAPK(p38丝裂原活化蛋白酶)水平的影响。方法:将12周龄雄性新西兰大白兔48只,随机数字表法将其分为6组:丹连消痤散高剂组、中剂组、低剂组、阳性对照组(克痤隐酮凝胶)、模型对照组、正常对照组,每组8只。除正常组外,其余各组Kligman法制备兔耳痤疮模型,并分别皮肤给药,每日1次,连续14 d。免疫组化法检测兔耳NF-κB、p38MAPK蛋白表达。结果:模型组兔耳NF-κB、p38MAPK蛋白表达较正常组明显增强(P <0. 01),与模型组比较,阳性对照组、高剂组、中剂组、低剂组兔耳NF-κB、蛋白表达明显减弱(P <0. 05)。正常组、模型组与各组间均有显著差异,差异有统计学意义(P <0. 05)。结论:丹连消痤散能够明显抑制痤疮模型兔耳NF-κB、p38MAPK表达,减轻炎症反应。
Objective: To observe the effects of Danlian Xiaocuo Powder on the expressions of NF -κB and p38MAPK in rabbits' ear with acne. Methods: 48 male New Zealand white rabbits aged 12 weeks,random number table method was used to divide them into six groups: Danlian Xiaocuo Powder high dose group, medium dose group ,low dose group, positive control group ( Clindamycin gel), model control group, normal control group, 8 rabbits in each group. Except the normal group, each group was modeled according to Kligman' s method and given skin medication,once a day for 14 days. NF-kappa B and p38MAPK protein expressions in rabbits' ears with acne were determined by immunohistochemistry. Results: The expressions of NF-κB and p38MAPK in rabbits' ears of model group was significantly higher than those of normal group (P < 0. 01). Compared with model group, the expressions of NF-κB and p38MAPK in rabbits' ears of positive control group, high dosage group, medium dosage group and low dosage group were decreased significantly (P < 0. 05). There were significant differences between normal group, model group and each medication group (P < 0. 05). Conclusion: Danlian Xiaocuo Powder can significantly inhibit the expressions of NF-κB and p38MAPK and reduce inflammation.
