仁和解酲汤对急性酒精中毒小鼠肝损伤干预作用的实验研究 被引量:4

Interventional Effect of Renhe Jiecheng Tang (仁和解酲汤) on Acute Alcoholic Liver Injury in Mice

摘要 目的:探讨仁和解酲汤对小鼠急性酒精中毒肝损伤的干预作用。方法:将32只雄性ICR小鼠分为正常组、模型组、阳性对照组(异甘草酸镁注射液腹腔注射)和中药组(仁和解酲汤灌胃) 4组,每组8只。实验7 d后处死小鼠取血清和肝组织进行生化指标的检测,光镜观察肝组织形态学变化和免疫组化方法检测NF-κB(nuclear factor-kappa B,核转录因子-κB)蛋白的表达。结果:与模型组相比,中药组和阳性药物组均能降低血清ALT、AST水平,增强肝组织中SOD、GSH-PX、ADH和ALDH活性,降低MDA的含量;病理学观察可见中药组和阳性药物组酒精性损伤后肝组织的结构改善;免疫组化显示两药物组肝组织中NF-κB蛋白的表达较模型组下降。结论:仁和解酲汤对急性酒精中毒小鼠的肝损伤具有明显的保护作用,其机制与增强抗氧化应激、降低NF-κB蛋白的表达相关。 Objective:To explore the protective effect of Renhe Jiecheng Tang on acute alcoholic liver injury in mice.Methods:32 male ICR mice were divided into normal group,model group( distillate spirit administration by gavage),positive drug group( Magnesium Isoglycyrrhizinate Injection administration by intraperitoneal injection) and Chinese medicine group( Renhe Jiecheng Tang administration by gavage),8 mice in each group,and liver tissue and its related biochemical markers( AST,ALT),morphological changes and protein expression of NF-κB were detected in the mice after 7 days of experiment.Results:Compared with the model group,the activities of SOD,ADH,GSH-PX and ALDH in liver tissues were increased,and the content of MDA was decreased.The histology changes of the liver in the Chinese medicine group were improved.Immunohistochemistry results showed the decrease of the expression of NF-κB.Conclusions:Renhe Jiecheng Tang has obvious protective effect on the liver injury in mice by relieving inflammation from down-regulating the expression of NF-κB.
