新型载盐酸四环素根尖充填材料的制备及其性能 被引量:1

Preparation and characterization of a new root-end filling material loaded with tetracycline hydrochloride

摘要 目的:探讨新型载盐酸四环素根尖充填材料(NRFM-THC)制备、理化性能、细胞相容性及其对粪肠球菌的抑制作用.方法:在新型根尖充填材料中添加质量分数为1%盐酸四环素制备NRFM-THC;采用傅立叶红外光谱、X-射线衍射对其表征,并检测其固化时间、抗压强度、抗稀散性和X射线阻射性能;MTT比色法检测其细胞相容性;琼脂糖扩散法及直接接触法检测其对粪肠球菌的抑制作用.结果:NRFM-THC主要成分为羟基磷灰石、羧酸钙盐及氧化锆,固化时间为(11.7±0.6)min,固化1 d及7 d组抗压强度分别为(43.9±4.5)MPa及(69.6±13.5)MPa,X射线阻射值等效于(3.6±0.2)mm纯铝,抗稀散性能良好等;细胞毒性分级为0级或Ⅰ级,符合GB/T 16886.5—2003标准;对粪肠球菌有抑制作用.结论:NRFM-THC具有良好的理化性能、细胞相容性及抑制粪肠球菌性能. Objective:To prepare a novel root-end filling material loaded with tetracycline hydrochloride(NRFM-THC),and explore its physicochemical properties,cellular biocompatibility,and antibacterial effect on Enterococcus faecalis.Methods:NRFM-THC was successfully fabricated by doping 1%tetracycline hydrochloride into a novel root-end filling material(NRFM).The chemical structure of the NRFM-THC was characterized with X-ray diffraction(XRD)and Fourier transform infrared spectrophotometry(FTIR).Furthermore,the physicochemical properties including setting time,compressive strength,washout resistance,and radiopacity were investigated to evaluate its possibility of clinic applications.The cellular biocompatibility of NRFM-THC was carried out by dimethyl-thiazol-diphenyl tetrazolium bromide(MTT)assay in L929 cells.Finally,the antibacterial activity of NRFM-THC against Enterococcus faecalis was investigated by using the agar diffusion test and direct contact test.Results:The main components of NRFM-THC were hydroxyapatite,calcium carboxylate,and zirconium dioxide.The setting time of NRFM-THC was(11.7±0.6)min.After 1 day and 7 days of continuous solidification,its compressive strength reached up to(43.9±4.5)MPa and(69.6±13.5)MPa respectively.The radiopacity of NRFM-THC was equivalent to that of aluminum with a thickness of(3.6±0.2)mm.In addition,the cytotoxicity scores of the NRFM-THC was grade 0 to I,which complied with the requirements of GB/T 16886.5-2003 standard on clinical medical materials.And the NRFM-THC displayed a significant antibacterial effect on Enterococcus faecalis.Conclusion:The excellent mechanical strength,outstanding biocompatibility,physicochemical properties,biocompatibility and antimicrobial activity that NRFM-THC can be a promising candidate as a promising candidate of root-end filling material.
