摘要
目的研究浙贝黄芩汤抗急性髓系白血病( acute myeloid leukemia,AML)部分作用机理。方法 21 例 AML 患者随机分为对照组 10 例、治疗组 11,对照组给予单纯化疗方案处理、治疗组予以浙贝黄芩汤联合化疗方案处理,比较治疗组治疗前后中医证侯积分变化,比较治疗组与对照组疗效,应用RT-PCR检测两组患者外周血中与张力蛋白同源的 10 号染色体缺失的磷酸酶基因( Phosphatase and tensin homolog deleted on ten chromosome,PTEN)、磷酸酰肌醇 3 激酶( Phosphatidyli-nosito1-3-Kinase,PI3K)表达。结果治疗组治疗后中医证候积分低于治疗前,差异具有统计学意义( P <0. 01);与对照组比,治疗组临床完全缓解、部分缓解、总有效率差异无统计学意义( P >0. 05),对照组总有效率稍低于治疗组;治疗前,对照组与治疗组 PTEN、PI3K mRNA 表达比较,差异无统计学意义( P >0. 05);治疗后,治疗组 PTEN mRNA 表达显著高于对照组,PI3K mRNA 表达显著低于对照组,差异具有统计学意义( P <0. 05)。结论上调 PTEN 表达和下调 PI3K 的表达可能是浙贝黄芩汤抗急性髓系白血病部分作用机理。
Objective To study the mechanism of Zhebei Huangqi Tang against acute myeloid leukemia ( AML). Methods A total of 21 patients with AML were randomly divided into the control group ( n =10) and the treatment group ( n =11). The control group was only treated with chemotherapy method while the treatment group was treated with Zhebei Huangqi Tang combined with chemotherapy method. The score changes of TCM syndrome before and after treatment in the treatment group were compared. The effi- cacy of the treatment group was compared with with the control group. Phosphatase and tensin homolog de- leted on ten chromosome,Phosphatidylinosito1-3-Kinase,PI3K expression in peripheral blood of two groups of patients were detected by RT-PCR. Results The scores of TCM syndromes in the treatment group were lower than those before treatment,and the differences were statistically significant ( P <0. 01). Compared with the control group,the differences in complete clinical remission ( CR),partial remission ( PR),and total efficiency ( OR) of the treatment group were not statistically significant ( P > 0. 05). The control group was slightly lower than the treatment group in OR. Before treatment,there were no significant differ- ences in PTEN and PI3K mRNA expression between the control group and the treatment group ( P > 0. 05). After treatment,the expression of PTEN mRNA was significantly higher than that of the control group,and the expression of PI3K mRNA was significantly lower than that of the control group ( P < 0. 05). Conclusion Up-regulation of PTEN expression and down-regulation of PI3K expression may be part of the mechanism of Zhebei Huangqi Tang on acute myeloid leukemia.
