摘要
目的研究新狼毒素A对Hep G2细胞糖酵解的影响。方法SRB法检测新狼毒素A对Hep G2细胞增殖的影响;台盼蓝拒染法检测细胞形态变化以及对细胞增殖的影响;试剂盒法检测新狼毒素A对Hep G2细胞三磷酸腺苷(ATP)含量的影响,试剂盒法检测新狼毒素A对细胞上清液中乳酸含量(LD)和葡萄糖含量的改变,以及新狼毒素A对细胞中己糖激酶(HK)、丙酮酸激酶(PK)和乳酸脱氢酶(LDH)活性的影响。结果新狼毒素A(0,20,40,80μg/m L)显著抑制Hep G2细胞的增殖且呈剂量依赖性;不同浓度的新狼毒素A作用48 h后,细胞中ATP含量逐渐降低,细胞上清液中葡萄糖含量逐渐增大和乳酸含量逐渐减小;糖酵解相关酶(PK和LDH)的活性也逐渐下降。结论新狼毒素A能够影响Hep G2细胞糖酵解水平,其机制可能与减少葡萄糖摄取,抑制糖酵解关键酶有关。
Objective To investigate the effect of Neochamaejasmin A on the glycolysis of Hep G2 Cells.Methods SRB assay was applied to assess the effect on proliferation of Hep G2 cells induced by Neochamaejasmin A.Typan blue exclusion was applied to observe cell morphology and effect on cell proliferation;The kit was applied to measure the content change of adenosine triphosphate(ATP)of Hep G2 cells induced by Neochamaejasmin A.The kits were employed in detecting the content changes of glucose and lactic acid content(LD)in the supernatant of Hep G2 cells induced by Neochamaejasmin A;The kits were applied to detect the activities of hexokinase(HK),pyruvate kinase(PK)and lactate dehydrogenase(LDH)in cells after treatment with Neochamaejasmin A.Results Neochamaejasmin A obviously suppressed the proliferation of Hep G2 cells in a dose-dependent manner.The content of ATP in Hep G2 cells was dlecreased,and the glucose in supernatant was increased and lactate in the supernatant was decreased in Hep G2 cells.Moreover,the activities of glycolytic related enzymes(PK and LDH)were reduced in Hep G2 cells treatment with Neochamaejasmin A.Conclusion Neochamaejasmin A can inhibit the level of glycolysis in Hep G2 cells,which may be related to the reduction of glucose uptake and inhibition of key enzymes in glycolysis.
