摘要
目的筛选乌苏里瓦韦抗炎作用的活性部位,并探讨其作用机制。方法将小鼠单核巨噬细胞RAW 264.7经脂多糖(LPS)诱导培养,建立炎症模型后,经不同浓度(25、50、100μg·mL^-1)的乌苏里瓦韦不同极性部位(石油醚部位、乙酸乙酯部位、正丁醇部位及水部位)处理,用试剂盒检测RAW 264.7细胞中炎症因子NO与PGE2的分泌水平。结果质量浓度为50、100μg·mL^-1的乙酸乙酯萃取部位、正丁醇萃取与水萃取部位均可显著抑制LPS诱导的RAW 264.7细胞炎症因子PGE2、NO的释放水平,且具有较好的量效关系。结论乌苏里瓦韦的乙酸乙醇、正丁醇与水萃取部位均为抗炎活性部位,其抗炎机制可能与影响炎症因子释放有关。
Objective To screen the anti-inflammatory fractions of Lepisorusussuriensis(Regel et Maack)Ching,and to investigate its mechanism.Methods Inflammation model was established by lipopolysaccharide(LPS)-induced micemononuclear macrophage RAW264.7 cells,and then treated with different concentrations(25,50,100 μg · mL^-1)of different polar fractions(petroleum ether,ethyl acetate,n-butanol and aqueous phase fractions)from Lepisorusussuriensis(Regel et Maack)Ching,and measured the secretion levels of I NO and PEG2.Results Ethyl acetate,n-butanol and aqueous phase fractions at concentrations of 50 and 100 μg ·mL^-1 significantly inhibited PGE2 and NO levels in LPS-induced RAW 264.7 cells,and showed a welldose-effect relationship.Conclusion The ethyl acetate,n-butanol and aqueous phase fractions may betheanti-inflammatory active site of Lepisorusussuriensis(Regel et Maack)Ching,and the anti-inflammatory mechanism may be related to the inhibition of generation of inflammatory mediators.
