光裸星虫提取物对膜性肾病模型的治疗作用及机制研究

摘要 目的探究光裸星虫提取物对膜性肾病模型大鼠的治疗作用及机制。方法选择SD健康雄性大鼠50只,选取16只作为对照组,剩下的34只做成膜性肾病模型,建模成功的有32只大鼠,将其分为模型组和实验组,每组16只大鼠。实验组大鼠给予光裸星虫提取物溶液灌胃治疗,模型组和对照组大鼠给予0. 9%氯化钠溶液。HE染色进行光镜下大鼠肾组织变化检查、免疫荧光法进行显微镜下大鼠肾组织观察、电镜下观察大鼠肾组织变化;化学发光分析法检测三酰甘油(triglyceride,TG)水平;电化学发光免疫分析检测血清总蛋白(total protein,TP)、白蛋白(albumin,ALB)、尿蛋白排泄量(urinary protein excretion,UPE)水平;二乙酰-肟法检测血清尿素氮(blood urea nitrogen,BUN)水平;免疫组化检测α-actinin-4和ZO-1蛋白表达。结果在实验组大鼠给予光裸星虫提取物治疗结束后,同时检测3组大鼠血清因子指标,对照组大鼠血清中TP、ALB的水平高于实验组高于模型组,差异有统计学意义(P<0.05);模型组大鼠血清中TG、BUB水平高于实验组高于对照组,差异有统计学意义(P<0.05)。对照组大鼠肾组织α-actinin-4蛋白水平低于实验组大鼠低于模型组大鼠,差异有统计学意义(P<0.05);对照组大鼠肾组织ZO-1蛋白水平高于于实验组大鼠高于模型组大鼠,差异有统计学意义(P<0.05)。结论光裸星虫提取物对膜性肾病模型大鼠的治疗效果较好,可以有效降低尿蛋白含量。 Objective To explore the the therapeutic effect and action mechanism of sipunculus nudus extract in treatment of membranous nephropathy in rats. Methods Fifty healthy male SD rats were enrolled in the study,in which,16 rats were enrolled as control groups,and the other 34 rats were made into membranous nephropathy models,finally,32 rat model with membranous nephropathy were successfully established,who were divided into model group(n = 16) and experimental group(n = 16). The rats in experimental group were given sipunculus nudus extract by gavage,however,the rats in model group and control group were given 0. 9% sodium chloride solution by gavage. HE staining was used to examine the changes of rat kidney tissue under light microscope,and immunofluorescence method was used to observe the changes of rat kidney tissue under microscope,and electron microscope was used to observe the changes of rat kidney tissue. Moreover the levels of triglyceride(TG) were detected by chemiluminescence analysis,and the levels of total protein(TP) and albumin were detected by electrochemiluminescence immunoassay. In addition the levels of albumin(ALB) and urinary protein excretion(UPE),blood urea nitrogen(BUN) and the expression levels of alpha-actinin-4 and ZO-1 protein were detected by diacetyl-oxime method and immunohistochemistry,respectively. Results At the end of treatment,the serum cytokines were detected in the three groups. The serum levels of TP and ALB in control group were significantly higher than those in experimental group,which in the latter were significantly higher than those in model group(P < 0. 05). The serum levels of TG and BUB in model group were significantly higher than those in experimental group,which in the latter were significantly higher than those in model group(P < 0. 05). Moreover the levels of actinin-4 protein in renal tissue of rats in control group were significantly lower than those in experimental group and those in model group(P < 0. 05). In addition the levels of ZO1 protein in renal tissues of rats in control group were significantly higher than those in experimental group,which in the latter were significantly higher than those in model group(P < 0. 05). Conclusion The sipunculus nudus extract has obvious therapeutic effects on membranous nephropathy in rat models,which can effectively reduce the content of urinary protein.
