盐酸戊乙奎醚对脓毒症大鼠急性肺损伤时炎性因子及中性粒细胞弹性蛋白酶表达的影响 被引量:3

摘要 目的探讨盐酸戊乙奎醚对脓毒症大鼠急性肺损伤肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、中性粒细胞弹性蛋白酶(NE)表达的影响。方法健康雄性SD大鼠60只,随机分为3组(n=20):假手术组(Sham组)、脓毒症组(CLP组)、盐酸戊乙奎醚组(PHC组)。采用盲肠结扎穿孔术(CLP)复制脓毒症大鼠模型。观察各组大鼠术后一般情况,观察造模后5d内3组动物存活率,同时于造模后术后12h,取血标本,并留取支气管肺泡灌洗液及肺组织进行检测。应用普通光镜及电镜检测各组肺组织的病理变化情况,同时计算肺湿/干重比(W/D)。采集血标本检测血气指标,采用ELISA法检测支气管肺泡灌洗液(BALF)中TNF-α、IL-6和NE的水平。结果与Sham组比较,CLP组、PHC组TNF-α、IL-6、NE表达升高(P<0.05);与CLP组比较,PHC组TNF-α、IL-6、NE表达降低(P<0.05)。结论盐酸戊乙奎醚能够有效降低大鼠体内TNF-α、IL-6和NE的水平,能显著减轻肺损伤的病理损害,对脓毒症大鼠的肺损伤有保护效果。 Objective To investigate the effects of penehyclidine hydrochloride on the expression of tumor necrosis factor-α,interleukin-6 and neutrophil elastase(NE) in acute lung injury of rats with sepsis. Methods Sixty healthy male Sprague-Dawley rats were randomly divided into 3 groups: sham operation group,sepsis group(CLP group) and penehyclidine hydrochloride group(PHC group),with 20 rats in each group. The rat models with sepsis were established by cecal ligation and puncture(CLP). The changes of general condition and the survival rate at 5 d after modeling were observed. Moreover the blood specimens were taken at 12 hours after modeling,and bronchoalveolar lavage fluid(BALF) and lung tissue samples were taken for blood gas analysis. The pathological changes of lung tissue were observed under light and transmission electron microscope,and the lung wet/dry weight ratio(W/D) was calculated. In addition the levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) and neutrophil elastase(NE) in BALF were measured by ELISA. Results As compared with those in sham operation group,the expression levels of TNF-α,IL-6 and NE were significantly increased in CLP group and PHC group(P < 0. 05). As compared with those in CLP group,the expression levels of TNF-α,IL-6 and NE were significantly decreased in PHC group(P < 0. 05). Conclusion Penehyclidine hydrochloride can effectively reduce the levels of TNF-α,IL-6 and neutrophil elastase(NE) in rats with sepsis,which can significantly relieve lung injury and has protective effects on acute lung injury in rates with sepsis.
