胃癌组织miR-30b和HOXA1的表达及与临床病理特征的关系 被引量:5

摘要 目的探讨胃癌组织miR-30b和HOXA1表达及其与临床病理特征的关系。方法选择胃癌患者142例,实时荧光逆转录法及免疫组化法测定miR-30b及HOXA1在胃癌组织(胃癌组)及正常胃组织(癌旁组)中的表达水平,分析其与临床病理参数和预后的关系。结果胃癌组MiR-30bmRNA表达水平低于癌旁组,HOXA1mRNA表达水平高于癌旁组(P<0.01);胃癌组MiR-30b蛋白表达评分低于癌旁组,HOXA1蛋白表达评分高于癌旁组(P<0.01);胃癌组MiR-30b蛋白表达阳性率低于癌旁组、HOXA1蛋白表达阳性率高于癌旁组(P<0.01);MiR-30b、HOXA1蛋白表达与年龄相关性不明显(P>0.05);与TMN分期、病理学分级、复发、浸润深度、淋巴血管间隙浸润、肿瘤最大径、淋巴结转移相关性明显,且TMN分期越高、病理学分期越高、有复发、浸润深度越深、有淋巴血管间隙浸润、肿瘤最大径≥5cm、有淋巴结转移,MiR-30b蛋白阳性表达率越低、HOXA1蛋白阳性表达率越高(P<0.01);MiR-30b阳性组3年生存率及生存期均明显高于MiR-30b阴性组,HOXA1阴性组3年生存率及生存期均明显高于HOXA1阳性组(P<0.01)。结论胃癌组织MiR-30b表达降低,HOXA1表达增加;MiR-30b在胃癌的发生发展过程中起抑制作用,HOXA1起促进作用;MiR-30b高表达、HOXA1低表达的胃癌患者能获得较好的预后。 Objective To investigate the expression levels of miR-30b and HOXA1 in gastric cancer tissues and their relationship with clinicopathological features. Methods A total of 142 patients with gastric cancer were enrolled in the study.The expression levels of miR-30b and HOXA1 in gastric cancer tissues (gastric cancer group) and normal gastric tissues (para-cancer tissue group) were detected by real-time fluorescence reverse transcription and immunohistochemistry,moreover the correlation between the expression levels of miR-30b and HOXA1 in gastric cancer tissues and clinicopathological features was analyzed. Results The expression levels of miR-30b mRNA in gastric cancer group were significantly lower than those in para-cancer tissue group,however,the expression levels of HOXA1 mRNA were significantly higher than those in para-cancer tissue group ( P <0.01).Moreover the expression scores of miR-30b protein in gastric cancer group were significantly lower than those in para-cancer tissue group,however,the expression scores of HOXA1 mRNA were significantly higher than those in para-cancer tissue group ( P <0.01). The expression positive rate of miR-30b protein in gastric cancer group was significantly lower than that in para-cancer tissue group,however,the expression positive rate of HOXA1 protein was significantly higher than that in para-cancer tissue group ( P <0.01).Moreover the expression levels of MiR-30b and HOXA1 protein were not correlated with patient’s age,however,which were closely correlated with TMN stage, pathological grade, recurrence, depth of invasion, lymphatic vascular infiltration, tumor maximum diameter (≥5cm) and lymph node metastasis ( P <0.01). The 3-year survival rate and survival time of in MiR-30b positive group were significantly higher than those in MiR-30b negative group. The 3-year survival rate and survival time in HOXA1 negative group were significantly higher than those in HOXA1 positive group ( P <0.01). Conclusion The expression levels of miR-30b in gastric cancer tissues are decreased,however, the expression levels of HOXA1 are increased.The miR-30b has inhibitory effects in pathogenesis and development of gastric cancer,however, HOXA1 has promotive effects.The patients with high expression of MiR-30b and low expression of HOXA1 are expected to obtain better prognosis.
