摘要
采用超临界CO2萃取山药皮中的batatasins-I和一些挥发性成分.采用正交设计法研究了静态萃取时间、温度、压力以及各种改性剂对batatasin-I产率的影响.用高效液相色谱法测定提取物中的batatasin-I含量.结果表明,随着静态萃取压力、萃取时间和萃取温度的增加,batatasin-I的产率增加.添加改性剂可明显提高batatasin-I的产率,在40℃下,在40MPa下萃取4h,batatasin-I的产率最高可达395.54μg/g.以15%丙酮为改性剂,经气相色谱结合电子电离质谱(GC/MS-EI)测定,从山药皮中提取的batatasin-I的纯度可达到总提取物的23%.此外,还检测到了20余种挥发性成分.
Batatasins-I and some volatile ingredients from peels of Dioscorea opposita Thunb were extracted using supercritical CO 2.The effects of the parameters such as the static extraction time,temperature,pressure,and various modifiers on the yield of batatasin-I were investigated by the orthogonal array design.Batatasin-I content of the extract was determined by high-performance liquid chromatography (HPLC).The results indicated that the yield of batatasin-I was increased by increasing the static extraction pressure,time and temperature.The addition of modifiers can obviously improve the yield of batatasin-Ι.The highest batatasin-I yield of 395.54 μg/g was obtained when the extraction was carried out for 4 h at 40 ℃ under 40 MPa.When 15% acetone was used as the modifier,the purity of batatasin-I extracted from Dioscorea opposita Thunb peels can achieve to 23% of the total extracts calculated by gas chromatography combined with electron ionization mass spectrometry (GC/MS-EI).In addition,more than 20 volatile ingredients in the extract were also detected.
