摘要 [目的]探讨不同产地药材黄芪中总黄酮及异黄酮含量的差异。[方法]采用HPLC法测定了16个不同产地黄芪样品中总黄酮和4种异黄酮(毛蕊异黄酮苷、芒柄花苷、毛蕊异黄酮、芒柄花素)的含量,比较其差异性,并采用主成分分析法进行分析。[结果]不同产地黄芪中总黄酮及4种异黄酮含量均存在差异,蒙古黄芪和膜荚黄芪中总黄酮和4种异黄酮含量差异不显著(P>0.05)。16份样品中有5份样品的毛蕊异黄酮葡萄糖苷的含量符合药典不少于0.020%的要求。主成分综合得分结果显示,山西代县得分最高,其次为山西广灵、山西浑源泽清岭,且均为蒙古黄芪。SIMCA-P主成分分析可将16个不同地区的药用黄芪分为3类。[结论]膜荚黄芪与蒙古黄芪均可作为药材黄芪的基原物种。不同产地药材黄芪中总黄酮及异黄酮含量差异明显。应选择有效成分含量符合国家要求的地区进行黄芪的区域化种植。 [Objective] Present study was conducted to explore the differences of the content of flavonoids and isoflavones in Astragalus samples from different habitats.[Methods] The contents of total flavonoids and four isoflavones (isoflavone glycosides,ononin,calycosin and fermononetin) of Astragalus from 16 different areas were measured and comparedby HPLC,and analyzed using principal component analysis method.[Results] The differences of the total content of flavonoids and four isoflavones were detected in the samples from different habitats,while they were not significantly different between species A.membranaceus and A.mongholicus .The content of the isoflavone glucoside of 5 species out of 16 matched the requirement of pharmacopoeia with no less than 0.02% content standard.The results of the main component comprehensive score analysis showed the highest scoreat Dai county,followed by Guangling county,and Zeqingling village of Hunyuan county of Shanxi province,and the radix Astragali grown in these areas were belonged to species A.mongholicus. Samples of Astragalus species from 16 different regions were classified into three categories with SIMCA-P Principal Component Analysis method.[Conclusion] The differences between the components of A.membranaceus and A.mongholicus were not statistically significant,and they could be used as the basic species of Radix Astragali .However,the total content of flavonoids and isoflavones of Astragalus in different inhabits were significantly different ( P <0.05).The production areas where the content of active ingredients meet the standard requirement should be considered when planting Radix Astragali .
