摘要
目的 研究苯酚提取法对A群脑膜炎球菌粗制多糖原液中细菌内毒素含量的影响.方法 用苯酚对A群脑膜炎球菌多糖原液进行提取,比较粗制多糖的不同溶解倍数、苯酚提取过程中的不同搅拌转数、提取次数、提取温度等因素对多糖细菌内毒素含量的影响.结果 粗制多糖采用4.5% 醋酸钠溶液以1:100质量体积比溶解后进行苯酚提取的多糖细菌内毒素含量最低.同时,综合考虑多糖原液中蛋白含量与多糖回收率的检定结果,确定苯酚提取的最佳条件为搅拌转数400 r/min,提取次数3次,温度13~15℃,可有效降低多糖原液中细菌内毒素的含量.结论 苯酚提取法可明显降低A群脑膜炎球菌粗制多糖原液中的细菌内毒素含量.
Objective To investigate the effect of phenol extraction on endotoxin content in group A meningococcal polysaccharide solution. Methods The group A meningococcal polysaccharides were extracted with phenol. The effects of different dissolution ratios, speed of agitator, number of phenol extraction times, and phenol extraction temperature on the endotoxin content of polysaccharides were compared. Results When the crude polysaccharide was dissolved by 4.5% sodium acetate solution at 1∶100 mass to volume ratio before extracted by phenol, the content of endotoxin in polysaccharides was the lowest. Taking into account the protein content and polysaccharide recovery rate, the phenol extraction speed of 400 r/min, three times extraction, extraction temperature of 13-15 ℃ were chosen. Conclusion The phenol extraction method can significantly reduce the endotoxin content of group A meningococcal polysaccharide.
