摘要
目的:研究miR-34a在小鼠膝骨关节炎模型中的作用。方法:建立小鼠膝骨关节炎模型,行HE染色,对膝关节软骨损伤进行Mankin评分,然后取小鼠膝关节软骨细胞进行培养,RT-PCR检测miR-34a的表达,western-blot检测凋亡相关因子bcl-2、MMP-13的蛋白表达。结果:小鼠膝骨关节炎模型中,Mankin评分升高,miR-34a的表达升高,MMP-13亦升高,而bcl-2的表达降低。结论:在小鼠膝骨关节炎模型中,过表达的miR-34a,导致MMP-13的表达升高以及bcl-2的表达降低,加速骨关节炎进程。这说明沉默miR-34a可能成为防止骨关节炎进程的新方法。
Objective: To investigate the role of miR-34a on the mice knee osteoarthritis. Methods: Mice knee osteoarthritis model was established by hematoxylin-eosin staining and the cartilage injury of knee joint was assessed with Mankin score to confirm the successful establishment of osteoarthritis model.RT-PCR was used to detect mRNA expression of miR-34a,and Western Blot was used to detect the expression of bcl-2 and MMP-13. Results: In the mice knee osteoarthritis model,Mankin score increased,expression of miR-34a and MMP-13 increased,while the expression of bcl-2 decreased. Conclusions: In mice knee osteoarthritis model,over-expression of miR-34a leads to an increase in MMP-13 expression and a decrease in bcl-2 expression,and promotes osteoarthritis progression.This suggests that silencing miR-34a may become a new method for the prevention and treatment of osteoarthritis.
