摘要 以丹参叶片为外植体,将其接种在MS基本培养基上,探索不同激素种类和浓度配比对丹参愈伤组织诱导、愈伤组织分化为芽的影响。结果表明,MS+1.0mg/L6-BA+2.0mg/L2,4-D是丹参叶片诱导愈伤的最佳培养基;MS+2.0mg/L6-BA+0.25mg/LNAA是诱导愈伤组织分化为芽的最佳培养基。单独使用6-BA不利于诱导芽,与一定浓度的NAA配合使用能高效率地诱导芽。 In this research,Salvia miltiorrhiza leaves as explants were inoculated on MS basic medium to explore the effects of different types and concentrations of hormones on callus induction and callus differentiation into buds of Salvia miltiorrhiza.Different concentrations of 6-BA and 2,4-D were added to MS solid medium to induce callus of Salvia miltiorrhiza leaves and the induction rate were calculated.After subculture of callus,the callus with good growth were selected to cultivate under different concentrations of 6-BA and NAA in MS solid medium,then calculated the germination number and germination rate of callus.The results showed that the best medium of callus induced was MS+1.0 mg/L 6-BA+2.0 mg/L 2,4-D by Salvia miltiorrhiza leaves,the optimum medium of callus differentiation was MS+2.0 mg/L 6-BA+0.25 mg/L NAA.The use of 6-BA alone is not conducive to inducing buds,and the use of induction buds with a certain concentration of NAA can be more efficient.This study will provide technical support for fast,efficient and stable reproduction of Salvia miltiorrhiza.
