摘要
目的:探讨檞皮素对非小细胞肺癌细胞miR-16和HOXA10表达的影响。方法:选择非小细胞肺癌患者44例,取其手术病理组织细胞常规培养,检测其中miR-16和HOXA10的表达;观察檞皮素对非小细胞肺癌细胞的抑制作用;探讨檞皮素对其细胞活性的影响。结果:非小细胞肺癌细胞中miR-16和HOXA10的表达水平随着患者的临床分期增加而升高(P<0.001)。在檞皮素作用后,非小细胞肺癌细胞中miR-16和HOXA10的表达水平在临床患者中均显著低于作用前(P<0.001),同时临床Ⅰ期-Ⅳ期细胞存活率均显著低于作用前(P<0.001),而凋亡率显著高于作用前(P<0.001)。结论:非小细胞肺癌细胞中miR-16和HOXA10的表达水平随着患者的临床分期增加而升高,而檞皮素能够有效抑制各个时期非小细胞肺癌细胞中miR-16和HOXA10的表达和存活率,增加凋亡率。
Objective:To investigate the effect of quercetin on the expressions of miR-16 and HOXA10 in non-small cell lung cancer cells.Methods:44 patients with non-small cell lung cancer were collected in this study.The pathological tissue cells from these patients were routinely cultured to detect the expressions of miR-16 and HOXA10,to observe the inhibition of quercetin on non-small cell lung cancer cells,to discuss effect of quercetin on cell viability.Results:The expressions of miR-16 and HOXA10 in non-small cell lung cancer cells were increased with the clinical stage of the patients ( P <0.001).After quercetin treatment,the expressions of miR-16 and HOXA10 in non-small cell lung cancer cells were significantly lower in clinical patients than before ( P <0.001),and the clinical stage I-IV cell survival rates were significantly lower than before ( P <0.001),the apoptotic rate was significantly higher than before ( P <0.001).Conclusion:The expressions of miR-16 and HOXA10 in non-small cell lung cancer cells were increase with the clinical stage of the patients.Quercetin can effectively inhibit the expressions of miR-16 and HOXA10 and cell survival rates in non-small cell lung cancer cells at various stages,and increase the rate of apoptosis.
