摘要
目的结直肠癌(Colorectal cancer,CRC)中肿瘤相关巨噬细胞(TAMs)CD68^+及CD163^+的检测及与基因表型的关系。方法免疫组化方法检测41例CRC肿物,癌旁5 cm(M5)、10cm(M10)。不典型增生(Atypicalhyperplasia,AH)组织CD68^+及CD163^+的表达,采用病理切片扫描仪细胞计数。对41例CRC肿物进行基因检测。结果 CRC患者CD68^+及CD163^+检测肿物组织高于癌旁5cm、10cm(P<0.01);肿物与不典型组织比较差异无统计学意义(P>0.05);癌旁组织M5与M10比较差异无统计学意义(P>0.05);回归分析提示CD163^+表达与K-ras基因突变有显著性关系(P<0.05)。CD68^+及CD163^+的表达与B-raf基因突变、MSS(微卫星稳定型)、淋巴结转移、脉管癌栓、神经侵犯无相关性(P>0.05)。结论CD68^+及CD163^+在结直肠癌肿物间质中高表达,并高于癌旁组织,与AH表达无相关性。CD163^+在肿物的表达与K-ras基因突变有相关性。
Objective To detect the expression of CD68^+ and CD163^+ in tumor-associated macrophages(TAMs) in colorectal cancer(CRC) and its relationship with gene phenotype. Methods Immunohistochemical method was used to detect the expression of CD68^+ and CD163^+ in 41 cases of CRC, paracancer 5 cm(M5), 10 cm(M10), and atypical hyperplasia(AH) tissues. Cell counts were performed using a pathological slice scanner. Gene detection was performed on 41 CRC tumors. Results The expression of CD68^+ and CD163^+ in CRC patients was higher than that of paracancer 5 cm and 10 cm tissues(P<0.01). There was no significant difference between tumor and atypical tissue(P>0.05). The difference was not statistically significant between paracancer M5 and M10 tissues(P>0.05). Regression analysis indicated that CD163^+ expression was significantly associated with K-ras gene mutation(P<0.05). The expression of CD68^+ and CD163^+ was not associated with B-raf gene mutation, MSS(microsatellite stable), lymph node metastasis, vascular tumor thrombus, and neurological invasion(P>0.05). Conclusion CD68^+ and CD163^+ are highly expressed in the interstitial of colorectal cancer, and higher than the adjacent tissues, and have no correlation with AH expression. The expression of CD163+ in the tumor is correlated with the K-ras gene mutation.
