摘要
目的探讨血清miR-19a与基质金属蛋白酶9(MMP-9)表达水平与骨肉瘤临床病理及预后的关系。方法选择2016年1月~2017年6月间我院收治的骨肉瘤患者81例作为研究对象,设为骨肉瘤组,同期选取在我院体检的健康人群35例作为对照组。比较两组受试者的血清miR-19a与MMP-9表达水平变化情况,分析骨肉瘤患者血清miR-19a与MMP-9表达水平与临床病理参数的关系,术后随访12个月,比较复发者和未复发者血清miR-19a与MMP-9表达水平变化情况。结果骨肉瘤组患者的血清miR-19a与MMP-9表达水平均高于对照组,差异有统计学意义(P<0.05)。血清miR-19a与MMP-9表达水平与骨肉瘤患者的年龄、性别、肿瘤大小无明显相关性(P>0.05);但TNM分期Ⅱb/Ⅲ者高于Ⅱa分期者,远处转移者高于未转移者,术后随访12个月,复发组的血清miR-19a与MMP-9表达水平均高于未复发组,差异有统计学意义(P<0.05)。结论骨肉瘤患者血清miR-19a与MMP-9表达水平明显升高,其可能参与了骨肉瘤的发生发展,术后对其检测在骨肉瘤患者的预后判断中具有一定价值。
Objective To investigate the relationship between serum miR-19a and matrix metalloproteinase-9(MMP-9) expression levels and clinicopathology and prognosis of osteosarcoma. Methods 81 patients with osteosarcoma who were admitted to our hospital from January 2016 to June 2017 were selected as the study subjects and were assigned to the osteosarcoma group. In the same period, 35 healthy people who received physical examination in our hospital were selected as the control group. The changes of serum miR-19a and MMP-9 expression levels in the two groups were compared. The relationship between serum miR-19a and MMP-9 expression levels and clinical pathological parameters in the patients with osteosarcoma was analyzed. The follow-up was carried out for 12 months after surgery to compare the changes of serum miR-19a and MMP-9 expression levels between the patients with recurrence and the ones without recurrence. Results The serum levels of miR-19a and MMP-9 in the osteosarcoma group were higher than those in the control group, and the differences were statistically significant(P<0.05). There was no significant correlation between serum miR-19a and MMP-9 expression levels and age, gender and tumor size of osteosarcoma patients(P>0.05);however, those with TNM stage Ⅱb/Ⅲ were higher than those with stage Ⅱa, and those with distant metastasis were higher than those without metastasis. After 12 months of postoperative follow-up, the expression levels of serum miR-19a and MMP-9 in the recurrent group were higher than those in the non-recurrent group, and the differences were statistically significant(P<0.05). Conclusion The expression levels of serum miR-19a and MMP-9 in osteosarcoma patients are significantly increased, which may be involved in the occurrence and development of osteosarcoma, and their postoperative determination has a certain value in the prognosis of patients with osteosarcoma.
