成骨生长肽衍生物抗去卵巢大鼠骨质疏松作用的实验研究

摘要 目的通过建立去卵巢大鼠骨质疏松模型来研究成骨生长肽衍生物对去卵巢大鼠骨质疏松的防治作用。方法选取3月龄未交配雌性健康SD大鼠48只,按体重随机分为6组,其中5组行双侧卵巢切除术,建立骨质疏松模型,1组行假手术,给药组分别给予一定剂量的ALEN、OPG、OPG1、OPG2。给药12周后,测定骨密度,血清中的钙、磷、碱性磷酸酶、骨钙素。结果模型组与假手术组比较,骨密度明显降低(P<0.05);各给药组及模型组与S组比较,骨密度降低(P<0.05),OGP1组与OGP2组比较,骨密度增加(P<0.05);模型组与假手术组比较,碱性磷酸酶活性明显升高(P<0.05);治疗组与模型组比较,碱性磷酸酶活性明显降低(P<0.05);与假手术组比较,其他各组骨钙素活性高(P<0.05);其他治疗组分别与阿伦磷酸钠治疗组比较,血清钙浓度升高(P<0.05);各治疗组分别与模型组比较,血清磷浓度增高(P<0.05)。结论成骨生长肽及其衍生物对去卵巢大鼠骨质疏松具有治疗作用,其中OGPA1对抗去卵巢大鼠骨质疏松的作用较好。 Objective To study the preventive and therapeutic effects of anti-osteoporosis of osteogenic growth peptide in ovariectomized rats by establishing osteoporosis model in ovariectomized rats. Methods A total of 48 3-month-old unmated female healthy SD rats were selected and randomly divided into 6 groups according to body weight. Five groups underwent bilateral ovariectomy to establish osteoporosis model. One group underwent sham operation. The drug-administered groups were given a certain dose of ALEN, OPG, OPG1, OPG2, respectively. After 12 weeks of administration, bone density, calcium, phosphorus, alkaline phosphatase, and osteocalcin in serum were measured. Results The bone density decreased significantly in the model group compared with that of the sham operation group(P<0.05). The bone mineral density decreased in each drug-administered group and the model group compared with that of the S group (P<0.05). The OGP1 group showed an increase in bone density compared with OGP2 group(P<0.05). Compared with that of the sham operation group, the alkaline phosphatase activity was significantly increased in the model group (P<0.05). The alkaline phosphatase activity was significantly lower in the treatment group than that of the model group (P<0.05). Compared with that of the sham operation group, the activity of osteocalcin was higher in other groups(P<0.05). The serum calcium concentration was higher in the other treatment groups than that in the alendronate treatment group(P<0.05). The serum phosphorus concentration was increased in each treatment group compared with that of the model group(P<0.05). Conclusion Osteogenic growth peptide and its derivatives have therapeutic effects on osteoporosis in ovariectomized rats. And OGPA1 has a better effect on osteoporosis in ovariectomized rats.
