摘要
目的:研究决银颗粒治疗银屑病的作用机理。方法:(1)采用鼠尾鳞片表皮模型(C57小鼠)考察决银颗粒对鳞片表皮角质层的影响。(2)采用小鼠阴道上皮模型(昆明种小鼠)考察决银颗粒对阴道上皮细胞有丝分裂的影响。结果:(1)对鳞片表皮角质层的影响:在光学显微镜下,决银颗粒低、中、高剂量组及复方青黛胶囊组小鼠尾部颗粒层细胞均高于生理盐水组,差异有统计学意义(P<0.05);决银颗粒高剂量组小鼠尾部颗粒层细胞高于决银颗粒低剂量组,差异有统计学意义(P<0.05)。决银颗粒低、中、高剂量组小鼠血清IL-2水平均低于生理盐水组,差异均有统计学意义(P<0.05或P<0.01);决银颗粒高剂量组小鼠血清IL-2水平低于复方青黛胶囊组,差异有统计学意义(P<0.05)。(2)对阴道上皮细胞有丝分裂的影响:有丝分裂细胞主要分布在基底层,决银颗粒高剂量组较少出现有丝分裂细胞,决银颗粒中剂量出现有丝分裂,决银颗粒低剂量较多出现有丝分裂。模型组出现较多有丝分裂细胞,模型组有丝分裂指数高于其余各组,差异有统计学意义(P<0.05)。决银颗粒低、中、高剂量组小鼠血清IL-10水平高于模型组,但差异无统计学意义(P>0.05);复方青黛胶囊组小鼠血清IL-10水平高于模型组,差异有统计学意义(P<0.05)。结论:决银颗粒能促进小鼠尾部颗粒层细胞的形成,降低血清IL-2水平,并随着剂量的增大而增强;对小鼠阴道上皮细胞有丝分裂有抑制作用,且随剂量的增加而增强。
Objective: To study the mechanism of treatment of psoriasis by Jue-yin Granule. Methods:(1)To observe the effect of Jue-yin Granule on the epidermis of scale through C57 mouse model.(2)To observe the effect of Jue-yin Granule on the mitotic of vaginal epithelial cells through Kunming mouse model. Results:(1)Effect on the cuticle of scaly epidermis: Under the optical microscope, the small, medium and high dose groups of Jue-yin Granule and the compound Qing-dai capsule group of mice showed higher granular cells in the tail than the saline group, and the difference was statistically significant(P<0.05);In the high dose group of Jue-yin Granule, the tail granular layer cells in mice were higher than the low dose group of Jue-yin Granule, and the difference was statistically significant(P<0.05);The low, medium and high dose of Jue-yin Granule group showed lower level of serum IL-2 was lower than saline group, and the difference was statistically significant(P<0.05 or P<0.01);The serum IL-2 level in the high-dose group of Jue-yin Granule was lower than those of the compound Qing-dai capsule group, and the difference was statistically significant(P<0.05).(2)Effect on mitosis of vaginal epithelial cells: Mitotic cells are mainly distributed in the basement, and mitotic cells appear less in the high-dose group of Jue-yin Granule;mitosis occurs in the dose group of Jue-yin Granule, and mitosis occurs in many of the low-dose groups of Jue-yin Granule. There were more mitotic cells in the model group, and the mitosis index in the model group was higher than the other groups, and the difference was statistically significant(P<0.05).The level of serum IL-10 in mice with low, medium and high dose of Jue-yin Granule was higher than that in the model group, but the difference was not statistically significant(P>0.05);The serum IL-10 level of compound Qing-dai capsule group was higher than that of model group, and the difference was statistically significant(P<0.05). Conclusion: Jue-yin Granules could promote the formation of empennage granular layer cells and decreased the serum level of IL-2. The effect of Jue-yin Granule could be enhanced with the growing doses. Jue-yin Granules could restrain the mitosis of vaginal epithelial cells in mice and the effect could be enhanced with the growing doses.
